Preliminary screening of antiproliferative activity of selected extracts of Centella asiatica

Centella asiatica is a tropical plant found in Madagascar and Asia particularly Indonesia, India and Malaysia. Its benefits for health have long been known in its native areas. Our elders use C. asiatica to cure a number of skin problems and heal small wounds. The present study examines the anti-proliferative effect of the crude extracts of C. asiatica as well as its partially purified fractions (CF) from silica gel column chromatographic procedures in vitro. The method used to verify the anti-proliferative activity was methylene blue staining assay by counting the number of living cells at the time of assay termination. A total of 11 accessions of C. asiatica methonolic crude extracts (MC) were subjected to a preliminary anti-proliferative bioassay screening against breast cancer (MCF-7), prostate cancer (DC145), ovarian cancer (CAOV-3), oral epidemoid (KB) and rybdosarcoma (RD) cell lines and compared with normal cell lines MDBK and VERO. Only, CA-03 (w Ip) had shown activity with IC50 values of 168, 178.5, 89.11 and 198 ug/ml on cell lines of KB, CaOV-3, MCF-7 and DUl45 respectively. No activity was detected for the rest of the extracts even at concentration as high as 200 ug/ml. The ethanolic crude extract (EC) of CA-03 (wp) when tested on CaOV-3, MCF-7 and DU 145 gave IC50 values at 99,125.8, 180 ug/ml and greater than 200 ug/ml respectively. The chloroform fraction (CF) showed good anti-proliferative activity on CaOV-3 with IC50 of 27.5 ug/ml. The other IC50 for CF were 36.3, 45 and 110 ug/ml on DU145, MCF-7and RD respectively. All the C. asiatica extracts were non-toxic to the normal cell lines.
