Bioautographic profile as standard reference for quantitative analysis of the efficacy of herbs as antiseptic and antioxidant

This study is designed to develop a standard reference for qualitative analysis on the efficacy of herbs as antiseptic and antioxidant with the aim to validate the authenticity of the herbal materials and quality control of their pharmaceutical herbal preparations. A method comprising a combination of bioautographic assay and thin layer chromatography (TLC) analysis was employd to detect the antioxidant and antiseptic biomarkers as well as to obtain the TLC profile. The herbal extracts were subjected to TLC analysis on silica gel plates and each separated compound was visualized by various detection methods.Various extract from three selected herbs, namely, piper betle L. (leaves).Ficus deltoidea Jack. (leaves) and Ruta angustifolia (L) Pers. (aerial parts) were studied. Antiseptic biomakers were were detected by using bioautographic agar overlay assay against selected standard bacterial and fungal strains, namely Staphylococcus aureus., Escherichia colli, Candida albicans, Microsporum canis and Triscophyton rubrum. The antioxidant biomarkers were determined using TLC and 2.2`-depheny1-2-picrylhydrazy1 (DPPH) staining assay. The complete biographic profile which included the antioxidant and antiseptic biomarkers with their TLC characteristic were used as standard reference for qualitative analysis on the efficacy of the herbs and and their efficacy and their herbal preparation. This method would also be applicable to other herbs for efficacy analysis on both of the medicinal properties tested here.
