Assessment of field resistance and identification of some Lr-genes in spring triticale samples | Оценка полевой устойчивости и идентификация некоторых Lr-генов у образцов яровой тритикале

Presented are the results of studying the field resistance of a large number of spring triticale samples to brown rust pathogen under the natural infectious background of the Moscow pathogen population in 2012-2015, as well as the identification of some effective Lr-genes in these samples. The study has shown a high level of field resistance of spring triticale to brown rust. During the growing season of 2012, 34 of the studied samples had no symptoms of brown rust, in 2013, there were 15 such samples. In the field conditions of 2014 and 2015, brown rust infestation was not visually observed in the studied samples of spring triticale. Using PCR markers, 176 of the studied samples were used to identify some effective Lr-genes. During identification, a fragment matching the size of the fragment amplified in the almost isogenic wheat line of Thatcher cultivar carrying the Lr19 gene was amplified in 24 spring triticale samples; another 14 samples were heterogeneous for the Lr19 gene. Also, 15 of the studied samples contained an amplified fragment matching the size with a fragment of an almost isogenic wheat line of Thatcher cultivar carrying the Lr25 gene. No gene markers Lr9, Lr12, Lr24, Lr28, Lr29, Lr25, and L47 were identified in the studied samples. When comparing the results of field stability assessment and PCR analysis, the authors found out that the presence in the genomes of the studied images of the Lr19 and Lr25 genes identified using DNA markers does not significantly affect the field stability of samples under study conditions. The study has proved the high resistance potential of spring triticale to brown rust pathogens, and a promising source material has been also determined this way. In particular, samples of spring triticale k-1200, k-1715, k-1716, PI495820, PI520484, Pamyati Merezhko, PRAG 554/1, Solovey Kharkovsky have shown resistance to brown rust for four years of research and can be recommended for use in breeding to a high level of field resistance to brown rust in the central regions of the Non-Chernozem zone. Meanwhile, the study identified an insignificant variability of the studied samples of spring triticale by effective resistance genes to the pathogen of brown rust, and proved the necessity of the introgression of Lr genes in the triticale genome.

Изучали полевую устойчивость (ПУ)206 образцов яровой тритикале (ЯТ) к возбудителю бурой ржавчины (БР) в условиях естественного инфекционного фона московской популяции патогена в 2012-2015 гг., а также результаты идентификации у этих образцов некоторых Lr-генов. Показан высокий уровень ПУЯТ к БР. В условиях вегетационного периода 2012 г. у 34 образцов из числа изученных отсутствовали симптомы БР, в 2013 г. таких образцов было 15. В полевых условиях 2014 и 2015 гг. поражение БР изученных образцов ЯТ визуально не проявлялось. При помощи ПЦР-маркеров у 176 образцов из числа изученных проведена идентификация некоторых эффективных Lr-генов. В ходе идентификации у 24 образцов ЯТ амплифицирован фрагмент, совпадающий по размеру с фрагментом, амплифицированным у почти изогенной линии пшеницы сорта Thatcher, несущей ген Lr19, еще 14 были гетерогенны по гену Lr19. У 15 образцов амплифицирован фрагмент, совпадающий по размеру с фрагментом у почти изогенной линии пшеницы сорта Thatcher, несущей ген Lr25. Маркеров генов Lr9, Lr12, Lr24, Lr28, Lr29, Lr25, L47 у изученных образцов идентифицировано не было. При сопоставлении результатов оценки ПУ и ПЦР-анализа установлено, что наличие в геномах изученных образов, идентифицированных при помощи ДНК-маркеров генов Lr19 и Lr25, не оказывает существенного влияния на ПУобразцов в условиях исследования. Показан высокий потенциал ЯТ по ПУ к возбудителю БР, выделен перспективный исходный материал. Образцы ЯТ к-1200, к-1715, к-1716, PI495820, PI520484, Памяти Мережко, ПРАГ 554/1, Соловей харьковский показали устойчивость к БР на протяжении 4 лет исследования, они могут быть рекомендованы для использования в селекции на высокий уровень ПУ к БР в центральных районах Нечерноземной зоны. Установлено незначительное разнообразие изученных образцов ЯТ по эффективным генам устойчивости к возбудителю БР и обоснована необходимость изучения генетики устойчивости, а также интрогрессии Lr-генов в геном ЯТ.
