The Effect of Fucoidan on the Gentamicin Induced Nephrotoxicity in Rats. Yüzüncü Yıl University, Health Science Institute, Department of Biochemistry, MSci. Thesis, Van 2014. Fucoidan is a sulphated polysaccharide derived from brown seaweed and used widely in the Far East due to its different phytotherapic effects. In this study, possible preventive or therapuetic effects of fuciodan, which has antioxidant properties and was administered together with gentamicin made up nephrotoxicity through giving gentamicin to the rats, have been searched. The rats used in this study were randomly grouped in three, (10 rats per group): control group, gentamicin group (80 mg/kg/day/i.p gentamicin was administered for 8 days), gentamicin+fucoidan group (80 mg/kg/day/i.p gentamicin for 8 days + fucoidan 100 mg/kg/day/ for 8 days were administered with intra-gastric gavage.) Blood and tissue samples were taken 24 hours after the 8 days trial period. Besides determining plasma, urea, creatinine, BUN, sodium, potassium, chloride levels in auto-analyser, glutathione in all the blood and plasma MDA and AOPP levels were analyzed spectrophotometrically. Additionally, histopathological examination of the kidney tissues was performed. The highest plasma urea, creatinine, BUN, potassium, AOPP and MDA levels were found in the gentamicin group. It was determined that while the levels of urea (p<0.01), creatinin (p<0.01), BUN (p<0.05), potassium (p<0.05), AOPP (p<0.01), MDA (p<0.01) in gentamicin+fucoidan group were decreasing with regard to those in gentamicin group, chloride levels were rising (p<0.05). Glutathione level was the highest in the control group; and statistical p<0.01 level of significance was observed between the control group and the other two groups. Consequently, looking at the positive changes it brought about in both the biochemical and histopathological data, it can be concluded that fucoidan administration together with gentamicin in nephrotoxicity created by gentamicine has renal protective quality and reduces nephrotoxic damage.
