Synthetic studies toward the insect pheromone lineatin | Syntesestudier mot insektferomonet lineatin

英语. The goal was to find an improved synthesis of the pheromone lineatin (1) that would include an intermediate compound that can also be used for a synthesis of another pheromone, grandisol (2). Since only small quantities are available from nature, they need to be produced in the laboratory in sufficient amounts that can be used in control and capturing of the producing insects. This study explores different strategies to access the core structure featured in lineatin and grandisol.The synthetic strategies were based on commercial available alchohols, 16 and 26, manipulating them into the allenes 13 and 23. Capitalizing on intramolecular [2+2] cycloaddition of an allene-ene motif as the pivotal transformation, the resulting [4+6]- bicyclic scaffold also serves as an access point to the natural product grandisol (2).Three strategies were attempted, but only the latter went as far as experimenting with the cyclization. Because time was limited, the cyclization was not optimized, only put through test runs. In this step it was interesting to compare different allen-enes 23a-c, but sadly none of them resulted in the bicyclic scaffold 22a-c.It was observed that some reactions went remarkably better with some experience, especially the making of allene 13. With more time available, it would be interesting to see if the allen-ene 12 would give better results than the allene-ene 23. It would also be interesting to compare the cyclisations of 12 and 23.Further studies of the cycloaddition would lead to synthesis of both lineatin (1) and grandisol (2), but due to time-limitations this was not thoroughly attempted in this study. Based on the suggested future work, it seems plausible that the synthesis of both pheromones could be completed using the new strategy.

Målet med oppgaven var å utvikle en ny og bedre syntese av feromonet lineatin (1) gjennom en strategi hvor et eller fler av mellomproduktene også kan benyttes i syntesen av feromonet grandisol (2). Feromonene er tilstede i så små mengder i naturen at de må produseres på laboratorier for at de skal kunne benyttes for kontroll og innfanging av insektene som produserer disse. Denne oppgaven tar for seg flere strategier for å oppnå kjernestrukturen for lineatin (1).Syntesestrategiene er basert på kommersielt tilgjengelige alkoholer, 16 og 26, som ble manipulert frem til allenene 13 og 23 . Nøkkeltrinnet i syntesen er en intramolekylær [2+2]-sykloaddisjon av allen-en som gir 3-oksabisyklo[4.2.0]oktan-skjelettet til lineatin (1). Dette mellomproduktet kan også benyttes i syntesen av grandisol (2).Det var tre strategier involvert i oppgaven, hvor kun tredje strategi nådde et punkt hvor forsøk på sykloaddisjon var mulig. Da tiden var begrenset ble det kun interessant å se hvorvidt syklisering fungerte. Det ble utført på tre forsøk med allen-en 23, men ingen gav ønsket bisyklisk produkt 22.Det ble erfart at noen reaksjoner ble bedre med erfaring, spesielt dannelse av allenet 13 i første strategi. Ved mer tid tilgjengelig kunne det vært interessant å se om allen-enet 12 hadde gitt bedre resultater enn allen-en 23, og om sykloaddisjonen hadde fungert bedre på dette allen-enet.Videre studier rundt trinnet vedrørende sykloaddisjon vil kunne medføre syntese av både lineatin og grandisol. På grunn av tidsbegrensinger ble ikke dette gjennomført i denne oppgaven, men studiene kom et godt stykke på vei. Arbeidet gjort i denne oppgaven og gjenværende trinn i eksisterende litteratur viser likevel at dette kan være en god syntesevei for lineatin (1) og grandisol (2).
