英语. Microorganisms known as producers of biological products and destructors of hydrocarbons are characterized by high spontaneous variability. It can lead to a change in their activity; therefore, during stabilizing selection, it is necessary to confirm periodically strain affiliation. The purpose of the research is assessment of the genetic diversity of bacteria of several genera for the selection of the most effective antagonist strains and destructor strains. The genotyping method used is based on the use of two endonuclease restrictions to cleave the genomic DNA of a bacterium. Present in the reaction mixture DNA polymerase (Taq) provides labeling of DNA fragments with biotinylated deoxycytosine triphosphate (Bio-dCTP). Genotyping of two closely related strains of Pectobacterium atrosepticum makes it possible to recognize about 50 DNA fragments, more than 20% of which are specific to one of the compared strains. 2 pairs of restriction endonucleases namely XbaI/DraI and XbaI/Eco24I were used; the results obtained indicate the same discriminative ability of enzymes when comparing P. atrosepticum strains D822 and G784 by genetic profiles. When genotyping Pseudomonas bacteria, the BcuI/Eco32I pair was optimal, for Bs, the first restrictase was — SgsI (39 cleavage sites), the second (reduces the size of the resulting DNA fragments) - Eco32I. The genetic profiles of microorganisms of the genera Pectobacterium and Pseudomonas, characterizing the individuality of each strain, were obtained by the DSST method. Significant genetic differences between Ps species fluorescens and Ps. marginalis were noted. At the same time, identical DNA fragments were identified, which indicates the generic phylogenetic proximity of these species. Based on the results of genotyping of Pseudomonas biodestructors, a collection of strains that utilize difficult-to-oxidize compounds, including heavy fractions of oil and polyaromatic hydrocarbons, has been formed, which makes it possible to form associations of destructor strains for the disposal of specific pollutants.

Микроорганизмы - продуценты биопрепаратов и деструкторы углеводородов характеризуются высокой спонтанной вариабельностью, что может приводить к изменению их активности, поэтому при стабилизирующем отборе необходимо периодически подтверждать их штаммовую принадлежность. Цель исследований - оценка генетического разнообразия бактерий нескольких родов для отбора наиболее эффективных штаммов-антагонистов и штаммов-деструкторов. Примененный метод генотипирования основан на использовании двух эндонуклеаз рестрикции для расщепления геномной ДНК бактерии. Присутствующая в реакционной смеси ДНК-полимераза (Taq) обеспечивает мечение фрагментов ДНК биотинилированным дезоксицитозинтрифосфатом (Bio-dCTP). Генотипирование двух близкородственных штаммов Pectobacterium atrosepticum позволяет распознать около 50 фрагментов ДНК, более 20% которых специфичны для одного из сравниваемых штаммов. Использовали 2 пары эндонуклеаз рестрикции - XbaI/DraI и XbaI/Eco24I; полученные результаты указывают на одинаковую дискриминационную способность ферментов при сопоставлении штаммов P. atrosepticum Д822 и Г784 по генетическим профилям. При генотипировании бактерий рода Pseudomonas оптимальной была пара BcuI/Eco32I, для Bs первая рестриктаза - SgsI (39 сайтов расщепления), вторая (уменьшает размер получаемых фрагментов ДНК) - Eco32I. Методом ДРИМ получены генетические профили микроорганизмов родов Pectobacterium и Pseudomonas, характеризующие индивидуальность каждого штамма. Отмечены существенные генетические различия между видами Ps. fluorescens и Ps. marginalis. При этом выявлены идентичные фрагменты ДНК, что указывает на родовую филогенетическую близость этих видов. По результатам генотипирования биодеструкторов рода Pseudomonas сформирована коллекция штаммов, утилизирующих трудноокисляемые соединения, включая тяжелые фракции нефти и полиароматические углеводороды, что позволяет составлять ассоциации штаммов-деструкторов для утилизации конкретных загрязнителей.
