Microrganismos associados às cigarrinhas vetoras de Xylella fastidiosa.

As cigarrinhas são insetos sugadores da família Cicadellidae que se alimentam principalmente nos vasos do xilema de plantas de citros, café e ameixa e transmitem a bactéria Xylella fastidiosa para estas plantas. Os principais sintomas na planta incluem amarelecimento e redução do tamanho de folhas, encurtamento de internódios, abscisão foliar e seca de ramos. Além de patógenos de plantas, os insetos se encontram associados a simbiontes, os quais podem ou não ser encontrados em especificas células nos insetos, proporcionando um benefício mutuo na maioria dos casos. O presente estudo teve como objetivo detectar a presença do fitopatógeno X. fastidiosa e dos simbiontes Candidatus Sulcia muelleri e Betaproteobacteria em três espécies de cigarrinhas, coletadas em plantas de café. Inicialmente foi feita a extração do DNA das amostras, depois PCR e por último a eletroforese para visualização dos resultados em gel de agarose. Para o simbionte S. muelleri obtivemos 45% de todas as cigarrinhas positivas, sendo que detectamos em 62% de Bucephalogonia xanthophis e em 28% da espécie Ferrariana trivittata. Já o simbionte Betaproteobacteria, foi detectado em 28% de todas as 3 espécies sendo 25% na espécie B. xanthophis, 21% em Acrogonia citrina e 53% em F. trivittata. A bactéria fitopatogênica X. fastidiosa foi detectada em um total de 17% dos indivíduos e apenas na espécie B. xanthophis. Este é o primeiro relato da existência de diferentes microrganismos em cigarrinhas da família Cicadellidae no Brasil, no entanto não sabemos qual é o seu papel na sobrevivência e no desenvolvimento desses insetos. Sharpshooters are plant-sucking insects of the Family Cicadellidae that feed mainly on xylem vessels of citrus, coffee and plum plants and transmit the bacterium Xylella fastidiosa to these plants. In addition to plant pathogens, insects are associated with symbionts, which may or may not be found in specific insect cells, providing a mutual benefit in most cases. The present study had as objective to detect the presence of the plant pathogen X. fastidiosa and the symbionts Candidatus S. muelleri and Betaproteobacteria in three species of sharpshooters collected in coffee plants. DNA was extracted from the samples, then PCR was conducted Finally the electrophoresis was performed to visualize the results on agarose gel. For the S. muelleri symbiont we obtained 45% of all positive insects, and detected in 62% B. xanthophis and in 28% of Ferrarian trivittata. Betaproteobacteria symbiont was detected in 28% of all three insect species, where 25% of which were detected in B. xanthophis, 21% of them in Acrogonia citrina and 53% in F. trivittata. The pathogenic bacterium X. fastidiosa was detected in a total of 17% of the individuals and only in the B. xanthophis species. This is the first report of the existence of different microorganisms in Cicadellidae in Brazil, but we do not know what their role is in the survival and development of these insects.
