Toxic effect of Ustilago scitaminea on sugarcane callus. 2. Culture filtrate action and comparison with the effects of U. maydis and kinetin

In experiments designed to investigate the toxic effect of Ustilago scitaminea on sugarcane tissue cultures, callus response to fungal culture filtrate appears similar to that recorded in simultaneous cultures. This shows that the toxic effect results from the production of fungal metabolites. The toxic effect can be modulated by varying either the amounts of filtrate added to or the number of parasitical colonies grown on the callus culture medium. Furthermore, demonstration of a similar callus response to a fungus which does not attack sugarcane, U. maydis, provides evidence that the in vitro toxicity is not specific of the U. scitaminea-sugarcane interaction. An inhibitory effect on sugarcane callus growth is also noticed with kinetin at concentrations starting from 10(-6) mol/l. However, various observations indicate that the excretion of cytokinin-like products by U. scitaminea cannot be the only explanation of the in vitro effect of the fungus
