Development and characterization of anthocyanins in Sangiovese berries and leaves from veraison to ripeness

We have made a through study of the Sangiovese vine, which represents 70-90% of the grape blend established by the regulations on the production of Chianti Classico and is of fundamental importance for all Tuscan wines, with regard to the single anthocyanins: cyanidin, delphinidin, petunidin, peonidin, malvidin (3-monoglucosides) in the berries. We followed the development of these anthocyanins "in vivo" from the herbaceous phase to ripening by means of HPLC, demonstrating that malvidin is obtained by transformation of cyanidin with the help of two enzymes: FH (flavonoid-hydroxylase), MT (methyl-transferase). Judging from previous "in vitro" studies, this seems to be the first pigment of the biological cycle. Lastly, we detected traces only of the acylate anthocyanins, a chemotaxonomical feature of the Sangiovese vine in Tuscany. It also became clear that the anthocyanins are not present in the leaves during the Sangiovese vegetative cycle, but that their traces appear near harvesting time

Il vitigno Sangiovese, che rappresenta il 70-90% dell'uvaggio stabilito dal disciplinare di produzione del Chianti classico e che e' fondamentale per tutti i vini toscani, e' stato studiato a fondo relativamente alla formazione delle singole antocianine: cianidina, delfinidina, petunidina, peonidina, malvidina (3-monoglucosidi) nelle bacche. Lo sviluppo delle antocianine e' stato seguito "in vivo" tramite HPLC, dalla fase erbacea alla maturazione, evidenziando che la malvidina, pigmento stabile, si forma ad opera della flavonoide-idrossilasi e delle metiltransferasi dalla cianidina, che da studi precedenti "in vitro" sembra essere il primo pigmento del ciclo biologico. Si e' rilevata infine la presenza, soltanto in tracce, delle antocianine acilate, il che definisce una caratteristica chemiotassonomica del Sangiovese in Toscana. Si e' messo in evidenza inoltre che le antocianine, durante il ciclo vegetativo del Sangiovese, sono assenti nelle foglie, salvo comparire in tracce all'epoca della vendemmia
