Comparison of in vitro solubilization with pig duodenal and faecal inocula of dietary components in coarse-ground barley

The in vitro solubilization was studied of dietary components in a coarse-ground barley employing duodenal digesta and faecal extracts from pigs as inoculum for the preparation of incubation media. There was a resemblance in the pattern of in vitro solubilization of dietary components between the two inocula, although the rates differed considerably. Dry matter was solubilized at a faster rate during the early stages of incubation with duodenal inoculum, probably due to the high content of hydrolysing enzymes. Starch was more readily solubilized than dry matter with both duodenal and faecal inocula. This was also true for crude protein during the early stages of incubations. However, in the intermediate phase with faecal inocula, crude protein was accumulated in the recovered residues, during which time dry matter and dietary fibre were extensively solubilized. The results indicate that, with coarse-ground feedstuffs either duodenal or faecal inocula could be used to study nutrient availability in the pig intestine, thus avoiding the need for cannulated animals. However, caution is needed, particularly with regard to crude protein disappearance.
