Effect on genotype, benzylamonopurine concentration, daylength and temperature on the formation of in vitro micro-tubers of potato (Solanum tuberosum L.)

The results of an experiment aimed at devising effective and practical means of producing micro-tubers from in vitro cultured single-node cuttings of potato are reported. Four cultivars were used: "Arsy", "Hermes", "Konst", and "Monalisa". Two culture media were tested, resulting from supplementing 2 and 4 mg/l of benzylaminopurine (BAP) to a medium containing the salt mixture of Murashige and Skoog, at a sucrose and agar concentration of 60 and 8 g/l respectively. The addition of activated charcoal to the medium did not prove useful. Two daylength (8 and 16 hours a day) and two temperature levels (20 and 26 degree C) were tested. The explants spontaneously produced a small amount of microtubers during the ageing, mainly depending on genotype and daylength. BAP presence proved to be determinant in increasing the rate of micro-tuberization, although its concentration had limited effect. Large differences were found among cultivars: "Monalisa" gave the best results both in number and weight of the micro-tubers, while the lower temperature increased their number. Interactive effects between genotype and temperature and between genotype and daylength on the weight of the micro-tubers were observed
