Histochemical and ultrastructural investigations on Longissimus Dorsi and Semimembranosus muscles in "Chianina breed" calves - at high or low feeding level

The investigations were performed in groups of Chianina Breed calves of 12 and 18 months of age respectively to verify if the different-high or low-nutritional level could influence the muscular fibres composition. We have analyzed muscles of calves six months old too. Specimens of cube cm 0.5 from Longissimus Dorsi (LD) and Semimembranosus (SM) muscles, quickly-frozen in isopentane and liquid nitrogen, were processed with myosinic ATPase, NADH-TR and M-alfa GPDH, to evidenziate alfa-R, beta-R and alfa-W muscular fibres. Similar specimens were fixed in glutaraldeyde and embedded in epoxy-resin. The ultrathin-sections were observed with transmission E.M. By means of image analysis system, some parameter of muscular fibres and myofibrils were calculated. Glycolytic-oxidative fibres prevail in LD muscle while glycolytic fibres are the most represented in SM muscle. We notice that muscles of animals reared with low-nutritional level present a percentage of oxidative fibres (beta-R) higher than that of calves reared with high-nutritional level
