Various explants of 30 economically important clones ofpoplars have been cultured on various media to compare theirgrowth rate and their regenerative potential. Additionally, a few clones were selected for controlled crossings. Explants ofparents and offspring have been grown to make a comparativestudy. Fresh leaves, callus and regenerated plantlets are beingused for isoenzyme and DNA analysis. The objective of the present investigation is threefold: 1. to compare the growth responses of different clones in vitro, 2. to regenerate plantlets for clonal multiplication, and 3. to investigate the role of extranuclear components in differentiation. A modified Murashige and Skoog's medium with supplementsproved most effective for the initiation of callus in a number of clones. The subculturable callus regenerated shoots on medium containing only NAA and BAP. One clone (Max 1) responded best on Whitehead and Giles medium. Of the various clones tried so far, two have yielded regeneration in more than 80% of the cultures. Interesting variation in response has been observed between the clones and also between the parents and the off-spring. For example one individual from one of the crosses regenerated shoots within two weeks, while the parents take much longer. To study the genetic stability of regenerated shoots, variousenzyme genotypes were scored in different tissues of Max 1 and from leaves of eight other clones. No variation was observed.
