Natural occurrence of aster yellows phytoplasmas on periwinkle in Southern Italy [Basilicata, Campania] and their molecular characterization

Naturally infected periwinkle (Catharanthus roseus) plants showing symptoms of yellowing, stunting, little leaf, virescence, phyllody and bunchy top appearance were observed in two different locations of Southern Italy. Plant-pathogenic phytoplasmas were detected in the diseased plants by fluorescence and electron microscopy and PCR-amplification of phytoplasma 16S rRNA gene sequences. They were transmitted by grafting and dodder to healthy periwinkle seedlings and characterized through fragment length polymorphism (RFLP) analysis of the amplified phytoplasma 16S RNA gene fragment by using AluI restriction endonuclease. Restriction site analysis data showed that phytoplasmas recovered from naturally infected periwinkles in Southern Italy belong to aster yellows cluster and are similar, on the basis of restriction profiles, to those affecting weeds and vegetable crops including tomato affected by big bud disease, recently investigated in several growing areas of Central and Southern Italy

Nella scorsa primavera-estate, piante di pervinca (Catharanthus roseus), naturalmente infette e mostranti sintomi di ingiallimento, nanismo, riduzione della lamina fogliare, virescenza, fillodia e aspetto affastellato in particolare nella parte apicale, sono state rinvenute in Italia meridionale in due differenti localita'. Fitoplasmi sono stati individuati in tali piante mediante l'ausilio della microscopia a fluorescenza ed elettronica e della reazione a catena della polimerasi (PCR), amplificando un frammento del gene che codifica la sintesi dell'RNA ribosomiale 16S di tali procarioti. Gli agenti patogeni sono stati trasmessi sperimentalmente mediante innesto e cuscuta a piante sane di pervinca e caratterizzati mediante l'analisi del polimorfismo della lunghezza dei frammenti di restrizione (RFLP) del frammento di DNA amplificato, impiegando l'enzima di AluI. L'analisi di restrizione ha evidenziato nei fitoplasmi studiati un profilo di restrizione tipico dell'agente del giallume dell'astro e simile a quello dei fitoplasmi rinvenuti recentemente su varie specie spontanee e ortive, ivi compreso il pomodoro affetto dalla malattia "big bud" in diverse aree coltivate dell'Italia centrale e meridionale
