Regeneration potential of tropical - subtropical - midaltitude and highland maize inbreds [Mexico]

Plant regeneration potential of 44 tropical and subtropical, 23 midaltitude, and eight highland maize inbred lines was evaluated during the first phase of a project directed at transforming CIMMYT maize (Zea mays L.) germplasm. Plants were grown at CIMMYT's experiment stations in El Batan and Tlaltizapan, Mexico. Immature embryos (1.0-1.5 mm) from selfed ears were harvested for establishing callus cultures. Basal N6 medium supplemented with 2 mg/l Dicamba (N6C1) proved better than other media tested for callus initiation and maintenance. When N6C1 medium was supplemented with silver nitrate, an increase in both embryogenic callus formation and regeneration potential was observed. Regenerable embryogenic calli were obtained from 50% of tropical and subtropical lines, 87% of midaltitude lines, and 75% of highland lines tested. Plant regeneration from embryogenic calli was achieved within the first 10 days after calli were transferred to N6 medium without auxins or to MS medium supplemented with IAA and 6-BAP. Complete plantlets were formed within 20-30 days and then transferred to the greenhouse for evaluation and seed production. Compared to the original plants, regenerated plants showed no major alterations in morphology or chromosome number. Successfull regeneration of these maize inbreds should provide the means to directly transform elite CIMMYT maize inbred germplasms

Nel corso della prima fase di un progetto finalizzato alla trasformazione di germoplasma di mais (Zea mays L.) del CIMMYT e' stato valutato il potenziale di rigenerazione delle piante di linee consanguinee, 44 tropicali e subtropicali, 23 di media altitudine e 8 di montagna. Le piante sono state coltivate alle stazioni sperimentali del CIMMYT di El Batan e di Tlaltizapan, Messico. Da queste, al fine di ottenere colture di calli, venivano raccolti embrioni immaturi (1.0-1.5 mm), prelevati da infiorescenze autofecondate. Dei substrati provati per la formazione e il mantenimento dei calli, il piu' adatto si e' dimostrato il substrato di base N6 arricchito con 2 mg/l di Dicamba (N6C1). Mediante l'aggiunta di nitrato d'argento al substrato N6C1 si otteneva un incremento sia della formazione del callo embriogenico, sia del potenziale di rigenerazione. Sono stati ottenuti calli embriogenici dal 50% delle linee tropicali e subtropicali, dall'87% delle linee di media altitudine e dal 75% delle linee di montagna studiate
