Detection of apple mosaic virus, apple chlorotic leaf spot virus and apple stem grooving virus in apple trees by enzyme-linked immunosorbent assay (elisa) using different plant tissues

The production and the use of virus-free propagating material is of great importance to prevent the spread of virus diseases, since most of the virus and virus-like diseases of apple trees are transmitted by infected propagating material. In Slovenia, a system for the production of virus-free plant material of fruit trees has not yet been implemented. The serological testing method enzyme-linked immunosorbent assay (ELISA) is needed for quick and efficient phytosanitary control and represents one of the essential parts of the project. Therefore, different plant tissues of infected apple trees taken at different growing stages have been studied for a detection of Apple Mosaic Virus (ApMV), Apple Chlorotic Leaf Spot Virus (ACLSV) and Apple Stem Grooving Virus (ASGV) using DAS ELISA method. Machine and manual sample preparation has been compared as well. Flower petals gave the best results in detection of ApMV, ACLSV and ASGV. Manual sample preparation was more efficient in detection of the three studied viruses.
