Determination of 2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyridine in a cigarette smoke condensate by HPLC with an electrode array detector

英语. A method based on HPLC with coulometric array detection for the determination of 2-amino-1-methyl -6-phenylimidazo[4,5-b]pyridine (PhIP) in cigarette smoke condensate has been developed. It consists of a two step purification on asilica cartridge and a preparative reversed phase column (RP-C18) followed by a separation on an analytical reversed phase column (RP-Select-B) using acetonitrile/methanol/glacial acetic acid/bidist. water 20:10:2:68, v:v:v:v, pH 5.1 as mobile phase. PhIP is detected in an electrode array detector (eight electrodes) between +580 and +700 mV. The average content of PhIP in cigarettes is 20.3 +/-1.2 ng/cigarette (n=6). The recovery was 75 +/-1.7% (n=6) and the detection limit in the cigarette condensate was found to be 1.3 microgram PhIP/l.

Nach einer Festphasenextraktion an einer SI-Saeule und einer praeparativen Chromatographie an einer "Reversed Phase" Saeule wurde PhIP von anderen Komponenten isokratisch (Acetonitril/Methanol/Eisessig/bidist. Wasser, 20:10:2:68, pH 5,1 v:v:v.v) an einer analytischen "Reversed Phase" Saeule getrennt und anschliessend in einem Elektroden Array Detektor (mit acht Elektroden) zwischen +580 und +700 mV detektiert. Der aus dem Kondensat ermittelte durchschnittliche Gehalt einer Zigarette betraegt 20,3 plus/minus 1,2ng (n=6).Die Ausbeute liegt bei 75,3 plus/minus1,7% (n=6) und die Nachweisgrenze bei 1,3 Mikrogramm PhIP/l.
