Influence of genotypes and cold pretreatment on the production of embryoids and their regenaration in tetraploid and hexaploid wheats [Triticum turgidum - Triticum aestivum]

Genetic variability and effect of cold pretreatment on androgenesis have been investigated in two tetraploid wheat (Triticum turgidum) genotypes, ENSAT-508 and Opale, and in one hexaploid wheat (T. aestivum) genotype, HD 112, in a controlled greenhouse. The experimental design was a randomized block with four replications. Each replication consisted of four pots with three plants. ENSAT-508 gave the best results in embryoid yield, but HD 112 produced the highest number of green plantlets per 100 embryoids. Cold pretreatment (7 days at 4 deg C +-1) reduced embryoid yield by 53% in ENSAT-508, by 68% in Opale and by 30% in HD 112. However, the overall rate of regeneration of green plantlets per 100 embryoids remained unchanged by cold pretreatment. The hexaploid genotype HD 112 showed an increased production of albino platlets with cold pretreatment. Thus cold pretreatment should not be considered as an efficient pretreatment for all genotypes and has to be investigated for each genotype, in order to determine its effects in anther culture response

Il disegno sperimentale era un blocco randomizzato con 4 replicazioni. Ogni replicazione era costituita da 4 vasi con 3 piante. ENSAT-508 ha fornito i migliori risultati nella produzione di embrioidi, ma HD112 ha prodotto il maggior numero di piantine verdi per 100 embrioidi.
