Localisation of antioxidant enzymes in maize leaves

The intercellular distribution of antioxidant enzymes was studied in maize leaves. Rapid separation of the mesophyll extract was used to minimise modifications in antioxidant status and composition due to extraction procedures. Enzyme and metabolite markers were used to determine the degree of purity of the mesophyll isolates. Ribulose 1,5 bisphosphate and ribulose 1,5 bisphosphate carboxylase/oxygenase were used as bundle sheath markers, while phosphoenopyruvate carboxylase was used as a mesophyll marker. Glutathione reductase and dehydroascorbate reductase were almost exclusively localised in the mesophyll tissue ascorbate peroxidase and superoxidase dismutase as well as glutathione and ascorbate were largely present in the bundle sheath cells.
