Molecular characterization of bacterial communities from phloem of elms affected by elm yellows [Ulmus]

英语. Phytoplasma, microorganisms belonging to the class of Mollicutes, are thought to be the causal agents of different diseases in many plant species. They reside in the phloem of infected host plants, but up to now it has not been possible to isolate and culture them in vitro. In the present report bacteria isolated from healthy and phytoplasma-infected elms trees were studied to ascertain whether the presence of phytoplasmas affected the composition, structure, and dynamics of resident bacterial populations. For this purpose more than 1,600 bacterial isolates, collected at different times from the roots and twigs of elm trees, were first grouped into more than 100 different clusters by restriction analysis of 16S rDNA (ARDRA). The high interspecific variability found was more pronounced in the microbial population from the roots than in that from the twigs, and in the population from the twigs of phytoplasma-infected trees than in that from the twigs of healthy trees. Strains belonging to the main ARDRA groups were identified by the determination of the 16S rDNA nucleotide sequence. RAPD analysis of about one hundred strains belonging to two main ARDRA groups revealed a high degree of intraspecific genetic variability, suggesting that the bacterial population had a non-clonal structure

Essi risiedono nel floema di piante ospiti infette, ma finora non e' stato possibile isolarli e coltivarli in vitro. Nel presente rapporto sono stati studiati i batteri isolati da alberi di olmo sani e infettati da fitoplasmi per verificare se la presenza di fitoplasmi influenzava la composizione, la struttura e la dinamica delle popolazioni batteriche residenti. A questo scopo, piu' di 1.600 isolati batterici, raccolti in tempi diversi dalle radici e dai rametti di alberi di olmo, sono stati innanzitutto raggruppati in piu' di 100 gruppi mediante analisi di restrizione del rDNA 16S (ARDRA). L'elevata variabilita' interspecifica riscontrata era piu' pronunciata nella popolazione microbica delle radici rispetto a quella derivante dai rametti e nella popolazione dei rametti degli alberi infettati da fitoplasmi rispetto a quella dei rametti degli alberi sani. I ceppi appartenenti ai principali gruppi ARDRA sono stati identificati mediante la determinazione della sequenza nucleotidica del rDNA 16S.L'analisi RAPD di circa cento ceppi appartenenti ai due principali gruppi ARDRA ha evidenziato un livello elevato di variabilita' genetica interspecifica, facendo ritenere che la popolazione batterica presentasse una struttura non clonale]
