Aureobasidium pullulans associated with russeting of apple and pear fruits [Malus pumila Mill. - Pyrus communis L.]

英语. Russeting of apple and pear fruit epidermal tissue, which often results in substantial losses to growers, can be attributed to several biotic and abiotic factors. In this study, we tried to confirm the etiological role of the dematiaceous hyphomycete Aureobasidium pullulans (De Bary) G. Arnaud in russet induction. This problem has been subject of previous studies in New York State. Strains of the fungus were isolated on semi-selective medium from fruit samples collected in different Italian fruit growing areas and were subsequently identified by means of scanning electron microscopy (SEM) and analysis of restriction fragment profiles from the products of a PCR-amplified variable region of ribosomal DNA (ribo-fingerprinting) including the intergenic spacer region ITS-1, the 5.8S gene and ITS-2. Phenotypic and molecular fingerprints characterizing the fungal strains isolated from russeted apple and pear fruits were indistinguishable from those of the authentic A. pullulans strains from different geographic origin, used as controls. The ability of our fungal isolates in causing fruit russet was assayed by repeated experimental inoculations on apple trees in the field. Factors and possible mechanisms involved in the onset of russeting are briefly discussed

意大利语. La rugginosita' del tessuto epidermico di mele e pere, non di rado all'origine di notevoli perdite economiche per il frutticoltore, puo' essere attribuita a diversi fattori biotici e abiotici. In questo studio, si e' cercato di confermare il ruolo eziologico dell'ifomicete dematiaceo Aureobasidium pullulans (De Bary) G. Arnaud nell'insorgenza di questa fitopatia, gia' oggetto di studio nello Stato di New York. Da campioni di frutti, raccolti in diverse aree frutticole italiane, sono stati isolati ceppi del fungo, successivamente identificati mediante microscopia elettronica a scansione (SEM) ed analisi dei profili di restrizione dei prodotti ottenuti da amplificazione genica (PCR) di una regione variabile di DNA ribosomiale (ribo-fingerprinting) comprendente lo spaziatore intergenico ITS-1, il gene 5,8S ed ITS-2. Le impronte fenotipiche e molecolari caratterizzanti i ceppi fungini isolati da epidermide rugginosa di mele e pere sono risultate indistinguibili da quelle di ceppi autentici di A. pullulans di diversa origine geografica usati come controllo. La capacita' dei nostri isolati di causare rugginosita' sui frutti e' stata saggiata mediante inoculazioni sperimentali ripetute di alberi di melo in campo. Infine, si discutono brevemente i fattori ed i possibili meccanismi implicati nell'insorgenza di rugginosita'
