Genetic diversity among Fusarium oxysporum f. sp. vasinfectum isolates revealed by UP-PCR and AFLP markers [Gossypium barbadense L. - Egypt]

英语. Fusarium wilt of cotton is responsible for significant yield losses worldwide. In this study we used UP-PCR and AFLP molecular marker techniques to evaluate and study the genetic similarity and phylogeny of 20 Fusarium oxysporum f. sp. vasinfectum isolates. On the basis of comparisons of genetic similarity matrices, AFLP markers were very similar to UP-PCR markers for estimating intra-specific genetic relationships. Dendrograms constructed with both AFLP and UP-PCR revealed that all accessions of F. oxysporum f. sp. vasinfectum isolates were grouped into the same cluster, in good agreement with the pathogenicity tests. In questo studio abbiamo utilizzato la UP-PCR e le tecniche dei marcatori molecolari AFLP per valutare e studiare la similarita' genetica e la filogenesi di 20 isolati di Fusarium oxysporum f. sp. vasinfectum. In base ai confronti fra le matrici di similarita' genetica, i marcatori AFLP risultavano molto simili ai marcatori UP-PCR per la valutazione delle relazioni genetiche intraspecifiche. I dendrogrammi, costruiti sia con AFLP, sia con UP-PCR, hanno messo in evidenza che tutte le accessioni degli isolati di F. oxysporum f. sp. vasinfectum erano raggruppate nel medesimo cluster, in buon accordo con i test di patogenicita'. La differenza principale fra le due tecniche consisteva nel fatto che i marcatori AFLP davano una miglior risoluzione rispetto agli UP-PCR ai fini della discriminazione fra Fusarium strettamente affini]
