Modeling the technology of genoidentification of plant raw materials for brewing from mixed multi-species crops | Моделирование технологии геноидентификации растительного сырья для пивоварения из смешанных разновидовых культур

The creation of DNA technologies for the identification of plant raw materials for breweries is in demand for practical implementation in the quality management system as part of the traceability of the initial stages of beer production. Along with barley, other crops are also used in brewing. The purpose of the study was to simulate the technology of genetic identification of plant raw materials for brewing consisting of a mixture of multi-species crops. The main emphasis was placed on the combination of barley with each of 7 species of vegetable raw. Restriction mapping of the aligned nucleotide sequences of the barley, wheat, rye, corn, rice, hop, oat, and buckwheat chloroplast DNA locus, as well as subsequent modeling of the corresponding PCR-RFLP profiles, was carried out using the NEBcutter V2.0 online tool. Mapping of the aligned nucleotide sequences (limited to control primers 1 and 2 from the Phire Plant Direct PCR Master Mix set) of the chloroplast DNA locus of 8 crop species over the identification-significant restriction sites of selected endonucleases (TaqI, AciI and BssSI) made it possible to calculate generated PCR-RFLP-profiles of vegetable raw materials for brewing, both for a single species and for mixed multi-species crops. By further in silico modeling of PCR-RFLP profiles, bioinformatic data of the corresponding restrictograms were generated, displayed in a graphical format. The generated PCR-RFLP profiles of TaqI-, AciI-, and BssSI-restrictograms of mixed species crops indicate the need to rely on a cumulative analysis of data obtained from 3 selected restriction endonucleases at once, since interpretation of an individual restriction endonuclease in the identification key is difficult due to similar patterns of electrophoretic separation of cleaved fragments of chloroplast DNA in different crop species.

Создание ДНК-технологий идентификации растительного сырья для пивоварения востребовано для практического внедрения в систему менеджмента качества в рамках прослеживаемости начальных этапов производства пива. Наряду с ячменем в пивоварении используют и другие с.-х. культуры. Цель исследования - моделировании технологии геноидентификации растительного сырья для пивоварения, состоящего из смеси разновидовых культур. Основной упор был сделан на комбинацию ячменя с каждым из 7 видов растительного сырья. Рестрикционное картирование выравненных нуклеотидных последовательностей локуса хлоропластной ДНК ячменя, пшеницы, ржи, кукурузы, риса, хмеля, овса и гречихи, а также последующее моделирование соответствующих ПЦР-ПДРФ-профилей проведено в онлайн-программе NEBcutter V2.0. Картирование выравненных нуклеотидных последовательностей (ограниченных контрольными праймерами 1 и 2 из набора Phire Plant Direct PCR Master Mix) локуса хлоропластной ДНК 8 видов с.-х. культур по идентификационно-значимым сайтам рестрикции подобранных эндонуклеаз (TaqI, AciI и BssSI) позволило рассчитать генерируемые ПЦР-ПДРФ-профили растительного сырья для пивоварения, как для отдельно взятого вида, так и для смешанных разновидовых культур. Дальнейшим in silico моделированием ПЦР-ПДРФ-профилей были сформированы биоинформационные данные соответствующих рестриктограмм, отображенных в графическом формате. Сгенерированные ПЦР-ПДРФ-профили TaqI-, AciI- и BssSI-рестриктограмм смешанных разновидовых культур свидетельствуют о необходимости опираться на совокупный анализ данных, полученных сразу от трех подобранных эндонуклеаз рестрикции, так как по отдельно взятой рестриктазе интерпретация в идентификационном ключе затруднительна ввиду схожих картин электрофоретического разделения расщепленных фрагментов хлоропластной ДНК у разных видов с.-х. культур.
