Identification of resistance genes in the framework of veterinary monitoring | Идентификация генов резистентности в рамках ветеринарного мониторинга

英语. The occurrence of antibiotic-resistant strains of microorganisms was monitored in 7 federal districts of Russia. Using real-time polymerase chain reaction (PCR-RT) 263 samples selected in 2021 and 2022 from clinically healthy animals were examined. The samples were droppings, feces, swabs from udders and from entities of animal and poultry management. The genetic determinants of resistance to 5 antibiotic classes were determined: tetracyclines (tetA, tetO, tetM); fluoroquinolones (qnrS, qnrB); cephalosporins, penicillins (CTX-M-1, CTX-M-9, CMY); olymyxins (mcr-1). In 95% (n=249) of the samples there were found genes associated with resistance to 2 classes of broad-spectrum antibiotics. In 60% (n=158) of the samples, resistance was revealed to tetA, in 52% (n=138) to tetM, in 48% (n=127) to tetO, in 42% (n=111) to qnrB, in 35% (n=92) - to qnrS, in 22% (n=58) - to CTX-M-1, in 13% (n=34) - to CTX-M-9, in 24% (n=63) - to CMY, in 19% (n=50) - to mcr-1. As to animal species, the genetic markers of antibiotic resistance can be arranged in ascending order as follows: sheep – horses – goats – cattle – pigs – poultry. This is consistent with the fact that antibiotics are used in the treatment of pigs and poultry more often than in the treatment of cattle. In samples taken from environmental entities, the detection rate of genetic resistance determinants was comparable to that in samples taken in poultry houses. Resistance genes were detected two to three times oftener in faecal samples than in environmental samples. The study has shown which resistance markers are common in samples from animals and the environment, and the relative amount and detection rate of these markers can serve as a parameter for assessing the risk of further spread of antibiotic resistance.

俄语. В 7 федеральных округах России был проведен мониторинг распространения резистентных к антибиотикам штаммов микроорганизмов. С помощью полимеразной цепной реакции в режиме реального времени (ПЦР-PВ) было исследовано 263 образца, отобранных в 2021 и 2022 гг. от клинически здоровых животных. Образцы представляли собой помет, фекалии, смывы с вымени и с объектов содержания животных и птицы. Определяли генетические детерминанты резистентности к 5 классам антибиотиков: тетрациклины (tetA, tetO, tetM); фторхинолоны (qnrS, qnrB); цефалоспорины, пенициллины (СТХ-М-1, СТХ-М-9, CMY); полимиксины (mcr-1). В 95% (n=249) образцов обнаружили гены, связанных с устойчивостью к 2-м классам антибиотиков широкого спектра действия. У 60% (n=158) образцов выявили устойчивость к tetA, 52% (n=138) - к tetM, 48% (n=127) - к tetO, 42% (n=111) - к qnrB, 35% (n=92) - к qnrS, 22% (n=58) - к CTX-M-1, 13% (n=34) - к CTX-M-9, 24% (n=63) - к CMY, 19% (n=50) - к mcr-1. По видам животных генетические маркеры резистентности к антибиотикам можно расположить по возрастанию в следующем порядке: овцы – лошади – козы – крупный рогатый скот – свиньи – с.-х. птица. Это согласуется с фактом, что антибиотики используются в лечении свиней и с.-х. птицы чаще, чем в лечении крупного рогатого скота. В образцах, отобранных из объектов внешней среды, частота выявления генетических детерминант резистентности была сопоставимой с таковой в образцах, взятых в птичниках. В образцах фекалий гены резистентности выявляли в два-три раза чаще, чем в образцах из окружающей среды. Исследование показало, какие маркеры резистентности распространены в образцах от животных и окружающей среды, а относительное количество и частота обнаружения данных маркеров могут служить параметром оценки риска дальнейшего распространения резистентности к антибиотикам.
