Induction of defense responses related to scavenging reactive oxygen species in Ampelopsis species inoculated with Rhizobium vitis

Reactive oxygen species (ROS) is an essential factor that mediates various defense mechanisms against foreign stresses in plants. This study evaluated the content of salicylic acid (SA) and hydrogen peroxide (H₂O₂) and expression of genes related to ROS scavenging and investigated the content of resveratrol and the expression of genes associated with the pathogenesisrelated protein and stilbene synthesis in the leaves of Ampelopsis strains inoculated with Rhizobium vitis. An accumulation of resveratrol and the expression of PR genes were induced in YG10042, YG10045, YG10064, YG11030, YG10075, and YG-Songni4 resistant to the pathogens. A correlation was observed between the ROS content and the gene expression patterns of catalase (CAT) in grapevines after pathogen inoculation. The H₂O₂ contents, expression of CAT and superoxide dismutase (SOD) gene, and SOD-like activity decreased in Ab2 B-8–2 after pathogen inoculation. The H₂O₂ content increased with increasing SA content and decreasing expression of CAT gene in YG10042 and YG10045. This suggests that H₂O₂-dependent pathways in the induction of the defense responses in YG10042 and YG10045 inoculated with R. vitis. In contrast, in YG10064, YG11030, YG10075, and YG-Songni4, in which the defense responses are induced via the H₂O₂ independent pathway, the expression of CAT and SOD genes increased, whereas the H₂O₂ content was decreased by the high expression of the CAT gene. Elucidating the roles of ROS accumulation and the expression of related genes would provide useful information in further studies on the mechanism of resistance to disease in Ampelopsis species.
