Studies on in vitro propagation of Anthyllis barba-jovis L

Initial in vitro culture establishment and first subculture of Anthyllis barba-jovis took place on solid Murashige and Skoog (MS) medium with 30 g L-1 sucrose supplemented with 1.0 mg L-1 BA. In a following subculture, the effect of cytokinin type, BA or ZEA, at 1.0 mg L-1, was examined in comparison with a hormone-free medium. Shoot production was obtained at 100% in all treatments. Higher shoot production (3.5 or 3.9 shoots explant-1) and longer shoots (2.4 or 2.1 cm) were observed on a medium containing BA or ZEA, respectively, in comparison to the hormone-free medium. Hyperhydrated shoots were formed at 25 or 45% when the medium contained BA or ZEA, respectively. MS medium with 1.0 mg L-1 BA is suggested for in vitro shoot production of A. barba-jovis. For in vitro rooting, microshoots (2.0 cm long) produced by sub-culturing were transferred to solid MS medium with 30 g L-1 sucrose, either without plant growth regulators or with 0.5 or 1.0 mg L-1 IBA, or to half-strength MS medium without plant growth regulators. Microshoots rooted at 90% when cultured on half-strength MS medium without plant growth regulators, while rooting was reduced in full-strength MS medium, and by the addition of IBA. Ex vitro acclimatization was highly successful (93%) on a mix of peat: perlite (1:1, v/v).
