Pathomorphological and haematological changes in experimentally induced lead toxicity in rats

The experiment was conducted on forty five Wistar rats which were divided into three groups. Rats of group I were provided with single dose of lead nitrate @ 600 mg/kg body weight. The group II rats were provided with lead nitrate @ 100 mg/kg body weight daily for 90 days by oral route. The group III rats were kept as control. The blood samples were collected on days 30, 60 and 90 for hematological and serobiochemical studies. Most of the clinical symptoms observed in the rats of group I on day 2ⁿᵈ and group II on day 50ᵗʰ post administration of lead nitrate were almost similar. The rats of group II revealed highly significant increase in lead concentration in blood, spleen, liver, kidneys and bones as compared to the values of animals in the group III. Pathomorphologically, morbid changes in the visceral organs were more marked in group I. Haematological observations revealed significant decrease in hemoglobin concentration, packed cell volume, total leucocyte count, differential leucocyte count, total erythrocyte count, mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration in chronic toxicity group. There were significant decrease in total protein, albumin and alkaline phosphatase values where as significant increase in glucose, serum urea, creatinine and aspartate aminotransferase values in chronic toxicity. Similarly, statistical analysis showed that bilirubin, alanine aminotransferase and serum cholesterol values were increased significantly in chronic toxicity group.
