Five acylation derivatives (1a–1e) were prepared by acylation of cytochalasin B (1), which was isolated from the marine-derived fungus Curvularia lunata. Compound 1 and its acylation derivatives 1a–1e were evaluated for their antiviral and cytotoxic activities in vitro. In particular, compound 1e exhibited potent antiviral activity against HSV and EV71 with IC₅₀ values of 29.5 and 21.4 μM, respectively, which were 4-fold and 11-fold stronger than that of the positive control, ribavirin.
