Responses of Arthrospira ZJU9000 to high bicarbonate concentration (HCO3−: 171.2 mM): How do biomass productivity and lipid content simultaneously increase?

The biomass productivity and lipid content of Arthrospira ZJU9000 were increased by 37.9% and 32.5% under high bicarbonate concentration (HCO₃⁻: 171.2 mM, 15 vol% CO₂ was bubbled into 300 ml culture medium at a flow rate of 30 ml/min). RNA sequencing results revealed that the pyruvate amount used for energy provision and fatty acid synthesis metabolic pathways was increased under high bicarbonate concentration (HCO₃⁻: 171.2 mM). The elongation and hydrogenation of carbon chains in the fatty acid synthesis pathway were enhanced, thereby promoting lipid accumulation. Proton transport from the matrix to the intermembrane was enhanced, subsequently increasing the potential energy among the membranes for ATP synthesis. The expression level of ATP synthase was 3.8 times higher than that of Arthrospira under air condition, resulting in the efficient conversion of ADP and phosphoric acid into ATP. High bicarbonate concentration (HCO₃⁻: 171.2 mM) enhanced the energy provision processes for biomass and lipid synthesis in microalgae, thereby simultaneously increasing the biomass productivity and the lipid content.
