The culture of adipose tissue explants in serum-free medium

Methods are described for culturing porcine and bovine adipose tissue explants in a serum-free medium for 48 h. With the appropriate mixture of hormones in the medium, tissue viability (as judged by capacity to maintain lipogenic and glucose-oxidative capacity) is maintained during the culture period. The maintenance of tissue viability is absolutely dependent on the presence of insulin (10 ng/ml) and hydrocortisone (50 ng/ml) in the medium. We have also found that a mixture of Ham's F12:alphaMEM (1:1 v/v) is equivalent to Medium 199 in maintaining adipose tissue viability. Because cell structure and function may be altered by the extracellular matrix, culturing adipose tissue explants may provide a more accurate perspective of tissue metabolism in vivo than can be gained by culturing isolated adipocytes. in summary, these methods provide a system that is quite useful for studying the endocrine regulation of adipose tissue metabolism where the hormonal effects are chronic in nature (e.g., due to changes in genomic expression or protein synthesis).
