In vitro morphogenic potential of differently aged Pinus radiata trees correlates with polyamines and DNA methylation levels

DNA methylation and polyamine content have been analysed in juvenile, mature vegetative and mature reproductive Pinus radiata trees. Juvenile individuals were characterised by a DNA methylation degree of 30% and a high ratio of free versus PCA (perchloric acid) soluble polyamine conjugates, while mature trees showed 60% methylcytosine and low ratio of free versus PCA-soluble polyamine conjugates. We propose that both indicators are related with the lost of morphogenic ability during pine ageing and so with the inability of mature trees to in vitro establishment through specific interactions. These interactions are discussed.
