A study of nonstarch carbohydrate in enzyme-treated hard red spring wheat gluten

Carbohydrate structure and amino acid composition of a nonstarch carbohydrate isolated from hard red spring wheat gluten were studied. Wheat gluten was fractionated using a series of enzyme treatments and gel filtration columns. Carbohydrate and protein of the isolated fraction were eluted coincidentally with Sephacryl S-200 and hydroxylapatite chromatography suggesting a covalent bond links between them. The molecular weight of the fraction was estimated to be 7000 daltons. Hydroxyproline, aspartic acid, threonine, and serine were found in the isolated nonstarch carbohydrates-peptides fraction. The ratio of arabinose, xylose, galactose, and glucose was 1:0.95:0.45:0.28, indicating the arabinoxylan/arabinogalactan structures in the isolated fraction. Arabinoxylan content was estimated at 64%. Methylated neutral and amino sugars were quantified and identified by gas chromatography and gas chromatography-mass spectrometry. Results indicated N-acetyl glucosamine-asparagine as a N-glycosidic linkage and xylose-threonine and arabinose-hydroxyproline as O-glycosidic linkage.
