Influence of parasitic gastroenteritis on dose-dependent pharmacokinetics and efficacy of albendazole in goats

Influence of parasitic gastroenteritis on kinetic disposition of albendazole was evaluated in parasitized goats (5000 epg) compared to that in naive goats (0 epg) following oral administration of the drug at 5.0, 7.5 and 10.0 mg kg⁻¹ body weight. Plasma levels of albendazole sulphoxide were significantly higher in goats with zero epg of faeces during 8–24 hrs. post-administration of albendazole at different dose rates. Pharmacokinetic analysis of data revealed significantly higher Cₘₐₓ, Tₘₐₓ and AUC in goats with zero epg of faeces, while significantly lower bioavailability of albendazole sulphoxide could be observed in goats with 5000 epg of faeces. The study indicated that the relative bioavailability of anthelmintically active moiety albendazole sulphoxide was considerably decreased in goats with 5000 epg at all the three dose rates. Extrapolation of data of albendazole sulphoxide in terms of drug-parasite contact revealed that the parasites are not exposed to the lethal toxic levels of the drug following administration of the drug at 5.0 mg kg⁻¹ body weight in highly parasitized goats but for a considerable lesser duration following oral administration of the drug at 7.5 and 10.0 mg kg⁻¹ body weight.
