Cycloprodigiosin uncouples H+ -pyrophosphatase of plant vacuolar membranes in the presence of chloride ion

Cycloprodigiosin hydrochloride (cPrG-HCl) at 10 nM uncoupled both H(+)-ATPase and H(+)-translocating pyrophosphatase in vacuolar membrane vesicles of mung bean, but did not inhibit the substrate hydrolysis. cPrG-HCl destroyed the transmembrane pH gradient only in the presence of Cl(-). The observations indicate that cPrG-HCl functions as a H(+)/Cl(-) symporter. Vesicle acidification was restored by SO4(2-) even in the presence of cPrG-HCl.
