Reisolation and enumeration of transformed Streptomyces strains from plant roots

A system was developed for the reisolation and enumeration of genetically-modified Streptomyces strains introduced into the root system by seed treatment. Two chromogenic markers, melanin production and actinorhodin production, were evaluated. Plasmids containing the genes for these marker systems and thiostrepton resistance were used to transform protoplasts of a streptomycin-resistant strain of Streptomyces griseus. A mineral salts medium containing starch as the carbon source and arginine as the nitrogen source (RASS) and amended with rifampicin, streptomycin and thiostrepton was found to be the most effective medium for reisolation and enumeration of the Streptomyces strains. Strains transformed with the actinorhodin marker system were shown to colonize wheat and alfalfa roots as well as the original strain.
