Phenolic and flavonoid content in roots and hypocotyls of resistant and susceptible bean plants (Phaseolus vulgaris L.) during early stage of colonization by Fusarium oxysporum f. sp. phaseoli

Fusarium wilt, caused by Fusarium oxysporum f. sp. phaseoli (Fop), is one of the most devastating vascular diseases of common bean worldwide. Although genetic resistance is the main strategy to disease control, few studies have been focused on defense mechanisms associated with bean resistance at early stages of colonization. Thus, this study aimed to evaluate the content of total phenolic and flavonoid production in susceptible (cv. Uirapuru) and resistant (UFSC-01) bean plants at 3 days after inoculation (dai), when the fungus is supposed to reach the vascular system. Phenolic and flavonoid contents were determined spectrophotometrically in root and hypocotyl extracts. At 3 dai, phenolic and flavonoid contents in hypocotyl tissues were, respectively, 2 and 5 times higher than those in roots. When compared to the susceptible bean plants, resistant genotype exhibited higher amounts of flavonoids, which were not affected by the inoculation. Fop caused an increase in phenolic content mostly in susceptible roots. Altogether, our results suggest that bean resistance to Fop is not related to phenolic and flavonoid accumulation in the beginning of vascular root colonization.
