Influencia de COMVIT B sobre la neurogénesis pre y postnatal de ratas wistar

Los requerimientos vitamínicos son cruciales durante las etapas de gestación para el desarrollo correcto del gestante. El complejo vitamínico Compvit-B® compuesto por tiamina, pridoxina y cianocobalamina, ha sido utilizado en tratamientos de neuritis y polineuritis de origen múltiple, neuropatías periféricas, ciáticas, parálisis facial, padecimientos con aumento de la utilización de energía como el estrés y el cáncer. Más recientemente este complejo vitamínico ha sido evaluado en el desarrollo de los procesos de memoria y aprendizaje en animales neonatos concluyendo su contribución a la ejecución exitosa de paradigmas cognitivos. Con el objetivo de evaluar el efecto neurogénico del Compvit-B® durante el desarrollo pre y postnatal del sistema nervioso se establecieron dos grupos experimentales con sus respectivos controles: Grupo Exp. 1: 4 ratas hembras gestante que recibieron una inyección ip. de 0.1ml del complejo vitamínico una vez al día durante los últimos 15 días del embarazo; Grupo Exp. 2: Neonatos de 15 días de nacidos, recibieron una inyección ip. de 0.1ml de complejo vitamínico una vez al día durante 15 días. Los grupos controles para ambos tratamientos recibieron una inyección ip. del vehículo utilizado para diluir la vitamina (agua estéril), bajo las mismas características metodológicas que sus grupos experimentales. Se evaluó la influencia del Compvit-B® en la memoria y el aprendizaje espacial, utilizando la prueba del Laberinto Acuático de Morris (LAM), posteriormente, los animales recibieron una inyección intracerebroventricular (I.C.V) de BrdU, que permitió el marcaje de las células en división. Finalmente se reveló histológicamente el marcaje de las células reactivas a BrdU y NeuN, confirmando la neurogénesis asociada a la aplicación del Compvit-B®. Los animales que recibieron Compvit-B® en estado embrionario tuvieron un mejor desempeño que su grupo control en la prueba LAM en la fase de transferencia; mientras que los neonatos evidenciaron diferencias significativas en la adquisición y la transferencia respecto a su control. En los resultados histológicos, hubo diferencias significativas en la neurogénesis únicamente en los neonatos tratados con Compvit-B®, con un mayor número de neuronas producidas en la zona hipocampal. Estos resultados evidencian, que la neurogénesis en neonatos mejora significativamente con la aplicación del Compvit-B®, y aunque no fue significativa la mejora de estos procesos neurogénicos por la estimulación vitamínica durante la fase embrionaria, el Compvit-B® posiblemente puede afectar positivamente la gestación; y el estado y desarrollo fisiológico de los neonatos.

ABSTRACT. The vitamin requirements are vital during the stages of pregnancy for the proper development of the fetus. The Compvit-B vitamin complex composed by vitamins B1, B6 and B12 has been used in the treatment of neuritis and multiple source polyneuritis, peripheral neuropathy, sciatica, facial paralysis, diseases with increased energy use as stress and cancer. Additional, this vitamin complex has been evaluated in the development of learning and memory processes in neonatal animals concluding his contribution to the successful implementation of cognitive paradigms. With the objective to evaluate the neurogenic effect of Compvit-B ® during nervous system development, it’s established two experimental groups with their respective controls: Exp Group 1: 4 pregnant female rats received an ip. Injection of 0.1 ml of the vitamin complex one once a day for the last 15 days of pregnancy; Exp Group 2: Neonates with 15 days old received an ip. Injection of 0.1ml multivitamin once a day for 15 days. Controls for both groups received an ip. Injection treatment, the vehicle used to dilute the vitamin (sterile water), under the same methodological features that their experimental groups. The influence of Compvit-B ® in memory and spatial learning was evaluated using the Morris water maze test (MWM), subsequently, the animals received an intracerebroventricular (ICV) injection of BrdU, this allowed to do monitoring to cell in division. Finally, the histochemistry against BrdU and NeuN showed positive cells. It confirmed the presence of neurogenesis associated with the application of Compvit-B®. The animals that received Compvit-B® in an embryonic state had better performance than the control group at the MWM test in the transfer phase, while neonates showed significant differences in the acquisition and transfer regarding their control. On the other hand, on the histological results, there were significant differences only in neurogenesis in neonates, with a larger number of neurons produced in the hippocampal area. These results show, the neurogenesis in neonates significantly improves with the application of Compvit-B®, and although it was significantly improve these neurogenic processes by stimulation of vitamin during the embryonic stage, the Compvit-B® could positively affect pregnancy, state and physiological development of infants.
