Determination of mycotoxins in a rumen fluid using ELISA method

The main goal of measurements was to verify the hypothesis, if is it possible to determine the content of mycotoxins in the rumen fluid by ELISA method. The impact of two different extraction reagents (water, methanol) for selected types of mycotoxins was tested during the experiment. Aflatoxins, ochratoxins, deoxynivalenol, fumonisin, T-2/HT-2 toxin, and zearalenon were selected for experiments. In this experiment, cannulated cows were used in accredited user equipment Agrovýzkum Rapotín s.r.o. Rumen fluid and the feed were taken from three cannulated cows over the period of four weeks (28. 6. – 27. 7. 2021). An important effect of the reagents used in extraction for analysis results was detected. For aflatoxins and fumonisin, distilled water was better extraction reagent in the samples of rumen fluid and hay. For ochratoxin and DON, methanol was a better extraction agent in the hay samples. For T-2/HT-2 toxin, a methanol was a better extracting agent in samples of rumen fluid and hay. For Zearalenon, both extraction agents were also suitable for samples of hay and rumen fluid. The experiment was also used as monitoring of mycotoxins in feeds in Agrovýzkum Rapotín s.r.o. All samples analysed were below the limit or recommended values set by EU or FDA. The highest amount of mycotoxins, 2.5 ppm of deoxynivalenol was measured in a hay third week of experiment.
