Coupling steroids with alkaline phosphatase for competitive immunoenzymatic assays in the plasma of various domestic mammals | Couplage d'un stéroïde à la phosphatase alcaline pour la réalisation de dosages immunoenzymatiques par compétition dans le plasma sanguin de différentes espèces de mammifères domestiques

英语. The concentrations of various steroids involved in animal reproduction and behavior, in particular progesterone, are analyzed by assays in the Endocrinology and Phenotyping Laboratory. Since progesterone is known to play a major role in reproduction in females, its concentration in plasma is analyzed by competitive immuno-enzymatic assay. With the halting of the sale of progesterone coupled with alkaline phosphatase (P4-Pal), the reactant essential for this competitive assay, we decided to develop a protocol for coupling steroids with alkaline phosphatase. Following this coupling for progesterone, and in the framework of the laboratory’s quality approach, the reliability, repeatability and reproducibility of the results were assessed. Since all these quality standards were satisfactorily met, we looked for the existence of correlations between the concentrations obtained in plasma samples with assays using the P4-Pal produced in the laboratory and those obtained using the P4-Pal initially marketed. For the 5 animal species studied mostly in the laboratory, the correlation coefficients r varied between 0.92 and 0.98. The protocol for coupling progesterone with alkaline phosphatase has been validated.

Les concentrations de divers stéroïdes impliqués dans la reproduction et les comportements des animaux, notamment la progestérone, sont mesurées par dosages au sein du laboratoire Phénotypage Endocrinologie. La progestérone ayant un rôle primordial dans la reproduction chez la femelle, sa concentration dans le plasma sanguin est mesurée par dosage immunoenzymatique par compétition. À l’arrêt de la commercialisation de la progestérone couplée à la phosphatase alcaline (P4-Pal), réactif essentiel à ce dosage par compétition, nous avons décidé de mettre au point un protocole de couplage de stéroïdes à la phosphatase alcaline. Suite à la réalisation de ce couplage pour la progestérone, et dans le cadre de la démarche qualité du laboratoire, la fiabilité, la répétabilité et la reproductibilité des résultats obtenus ont été évaluées. Tous ces critères de qualité étant satisfaisants, nous avons étudié les corrélations existant entre les concentrations obtenues dans des échantillons plasmatiques par dosages utilisant la P4-Pal produite au laboratoire et celles obtenues avec la P4-Pal initialement commercialisée. Pour les 5 espèces animales étudiées majoritairement au laboratoire, les coefficients de corrélation r variaient entre 0,92 et 0,98. Le protocole de couplage de la progestérone à la phosphatase alcaline est aujourd’hui validé.
