Nitric Oxide (NO) content and expression of genes involved in myogenesis in embryonal tissues of chickens (Gallus gallus domesticus L.) | Содержание оксида азота (NO) и экспрессия генов, ответственных за миогенез, в эмбриональных тканях кур (Gallus gallus domesticus L.)

英语. Nitric oxide is known to be involved in myogenesis in fowls. The work purpose was to compare changes in the NO content in the tissues of meat and egg chicken embryos with changes in the expression of a number of genes involved in myogenesis, in order to elucidate the mechanisms of the possible epigenetic effect of NO as a factor in the regulation of embryonic development. For the study, embryos of chickens of the Hisex White egg cross and the mini-meat breed (line A77, group 2), characterized by high and low intensity of embryonic NO oxidation, respectively, were used. In tissues of the embryo homogenate on day 6 and in homogenates of chest and thigh muscles on day 14, the expression of 7 genes involved in myogenesis or influencing it was assessed. During incubation, the concentration of embryonic NO and the intensity of its oxidation was determined in comparison with transcriptional activity of myogenesis genes as influenced by in ova administered nitroarginine (NA), a NO synthesis blocker, and green light (the factor that intensifies the oxidation of embryonic NO to nitrate but does not affects the intensity of NO synthesis). NA administered before incubation, on day 6, led to a decrease of 70% in the concentration of NO donor compounds in the embryo homogenate and increased the expression of the MyoD1, Myog and Mef 2c genes in Hisex White egg cross embryos. In the mini-meat chickens (line A77, group 2), which are characterized by a low level of deposited NO, the same trend occurred (a decrease in the concentration of donor compounds and an increase in the expression of the MyoD1, Myog and Mef 2c genes), but the difference between the native and treated embryos was less pronounced. The use of green light during incubation also contributed to an increase in the expression of the MyoD1, Myog and Mef 2c genes on day 6 of incubation.

Цель работы заключалась в сопоставлении изменения содержания NO в тканях эмбрионов кур мясного и яичного направлений продуктивности с изменением экспрессии ряда генов, вовлеченных в миогенез, для выяснения механизмов возможного эпигенетического эффекта NO как фактора регуляции эмбрионального развития. Для исследования были взяты эмбрионы кур яичного кросса Hisex White и породы мини-мясная (линия А77, группа 2), характеризующиеся соответственно высокой и низкой интенсивностью окисления эмбрионального NO. В тканях гомогената эмбриона на 6-е сут и в гомогенатах грудных мышц и мышц бедра на 14-е сут оценивали экспрессию семи генов, участвующих в миогенезе или влияющих на него. При инкубации определяли концентрацию эмбрионального NO и интенсивность его окисления в сопоставлении с транскрипционной активностью генов миогенеза при применении in ovo нитроаргинина (НА, блокатор синтеза NO) и зеленого света (фактор, способствующий интенсификации окисления эмбрионального NO до нитрата, но не влияющий на интенсивность синтеза NO). Введение НА перед закладкой на инкубацию на 6-е сут приводило к снижению концентрации соединений-доноров NO в гомогенате эмбриона на 70% и повышало экспрессию генов МyoD1, Myog и Mef 2c в эмбрионах кур яичного кросса Hisex White. У породы мини-мясных кур (линия А77, группа 2), для которых характерно низкое содержание депонированного NO, имела место та же тенденция (снижение концентрации соединений-доноров и повышение экспрессии генов МyoD1, Myog и Mef 2c), но разница между контролем и опытом была менее выражена. Использование зеленого света во время инкубации также способствовало повышению экспрессии генов МyoD1, Myog и Mef 2c на 6-е сут инкубации.
