Obtaining of callus cultures and induction of morphogenesis in Lavandula angustifolia Mill.: influence of cultivar, explant type and culture medium | Получение каллусных культур и индукция морфогенеза у Lavandula angustifolia Mill.: влияние сорта, типа экспланта и питательной среды

英语. The development of many biotechnological methods is based on optimization of exogenous and endogenous factors that limit the processes of plant regeneration from callus cultures. This is relevant for lavender too, which is one of the main essential oil plants grown in the south of Russia. The purpose of the work was to study the influence of the cultivar, hormonal composition of the medium and explant type on the induction of callus and morphogenesis in vitro in Lavandula angustifolia Mill. Various explants (leaf, stem and bud segments) of 3 cultivars (Stepnaya, Vdala, Merkuriy) cultivated on Murashige and Skoog (MS) medium added with alpha-naphthylacetic acid (NAA), 6-benzylaminopurine (BAP), kinetin (Kin), thidiazuron (TDZ) were introduced into the isolated culture. As found non-morphogenic or morphogenic (with buds and leaf primordia) callus was formed from all types of explants. The greatest ability to induce primary morphogenic callus was revealed for buds, which not only had a higher frequency of its formation (up to 55.3% of the number of explants), but also a wider range of media on which it was formed. The maximum morphogenic callus frequency when introduced in vitro was observed in Stepnaya (up to 14.3–55.3%), and the minimum – in Merkuriy (up to 2.5–30.0%). In the 1st passage, an advantage of using buds as explants, as well as the Stepnaya cultivar was also revealed. To induce indirect morphogenesis for all studied explants types, the efficiency of cultivation in 0–1 passage on MS medium added with 0.5 mg/L TDZ and 0.5 mg/L BAP was established. In the 1st passage the frequency of morphogenic callus formation on this medium reached 75.0–93.3% depending on a cultivar and explant. The feasibility of cultivating calli on induction media during 2 passages (up to 60–70 days) to identify their morphogenic potential is shown. The established optimal combinations of factors make it possible to improve the protocol for plant regeneration from lavender callus cultures.

俄语. Разработка многих биотехнологических методов основана на оптимизации экзогенных и эндогенных факторов, лимитирующих процессы регенерации растений из каллусных культур. Это актуально и для лаванды - одного из основных выращиваемых на юге России эфиромасличных растений. Цель работы – исследование влияния сорта, гормонального состава питательной среды и типа экспланта на индукцию каллусо- и морфогенеза in vitro у Lavandula angustifolia Mill. В изолированную культуру вводили различные экспланты (сегменты листа, стебля и почек) трех сортов (Степная, Вдала, Меркурий), которые культивировали на питательной среде Мурасиге и Скуга (МС) с добавлением альфа-нафтилуксусной кислоты (НУК), 6-бензиламинопурина (БАП), кинетина (Кин), тидиазурона (ТДЗ). Установлено, что из всех типов эксплантов формировался неморфогенный или морфогенный (с почками и зачатками листьев) каллус. Наибольшая способность к индукции первичного морфогенного каллуса выявлена для почек, у которых не только была выше частота его образования (до 55,3% от числа эксплантов), но и шире спектр сред, на которых он формировался. Максимальная частота морфогенного каллуса при введении in vitro отмечена у сорта Степная (до 14,3–55,3%), а минимальная – у сорта Меркурий (до 2,5–30,0%). В первом пассаже также выявлено преимущество использования в качестве эксплантов почек, а также сорта Степная. Для индукции непрямого морфогенеза у всех изученных типов эксплантов установлена эффективность культивирования в 0–1 пассаже на питательной среде МС с добавлением 0,5 мг/л ТДЗ и 0,5 мг/л БАП. В первом пассаже частота формирования морфогенного каллуса на этой среде достигала 75,0–93,3% в зависимости от сорта и экспланта. Показана целесообразность культивирования каллусов на индукционных средах в течение двух пассажей (до 60–70 сут.) для выявления их морфогенного потенциала. Установленные оптимальные сочетания факторов позволяют усовершенствовать протокол регенерации растений из каллусных культур лаванды.
