Alterações fisiológicas e bioquímicas em plântulas de soja em resposta à impedância mecânica do substrato e ao vigor de sementes | Physiological and biochemical changes in soybean seedlings in response to mechanical impedance of the substrate and seed vigour

A impedância mecânica é considerada uma preocupação importante da agricultura moderna. O estabelecimento das plantas no campo depende de boa emergência e crescimento das plântulas, além de sementes vigorosas. Todavia, carece de mais pesquisas visando elucidar os efeitos do impedimento físico do solo sobre o crescimento de plântulas de soja, bem como a interferência do vigor das sementes na tolerância à compactação. Além disto, é necessário o desenvolvimento de metodologias mais confiáveis para o estudo da impedância mecânica e das alterações nas plantas provocadas por este estresse. Dessa forma, os objetivos desse trabalho foram: i) desenvolver metodologia para o estudo da impedância mecânica na emergência e crescimento inicial de plântulas de soja; ii) avaliar as alterações fisiológicas e bioquímicas em plântulas em função do aumento da impedância mecânica do substrato; iii) avaliar o efeito do vigor das sementes na tolerância das plântulas ao aumento da impedância mecânica do substrato. Foi desenvolvido um sistema que pode ser utilizado para a avaliação das respostas de sementes e plântulas ao impedimento físico durante a germinação, emergência e crescimento de plântulas. Para validar o sistema foram utilizadas sementes de quatro cultivares de soja, que foram submetidas à emergência sob diferentes níveis de impedância mecânica do substrato (0,15, 0,60 e 0,87 MPa). As plântulas foram avaliadas quanto às características morfológicas. Para avaliar os efeitos do aumento da impedância mecânica do substrato na emergência e no crescimento inicial de plântulas de soja oriundas de sementes com diferentes níveis de vigor, bem como de características bioquímicas (produção de etileno, concentração de proteína e atividades de enzimas do sistema antioxidante) das plântulas, foram utilizadas sementes de duas cultivares sendo dois lotes de cada uma, com dois níveis de vigor (alto e baixo). As sementes foram submetidas durante a germinação e emergência das plântulas à diferentes níveis de resistência à penetração (0,15, 0,60 e 0,87 MPa) utilizando o sistema desenvolvido. Foi avaliado diariamente o número de plântulas emergidas. Após sete dias, as plântulas foram coletadas e submetidas à avaliação de parâmetros de crescimento (comprimento, diâmetro radicular e massa seca), além das avaliações bioquímicas. Os dados foram utilizados para caracterizar os efeitos da impedância mecânica nas características morfológicas, fisiológicas e bioquímicas das plântulas. A metodologia é promissora para fins de pesquisa em melhoramento de plantas visando verificar a variabilidade genotípica e selecionar genótipos mais tolerantes à compactação do solo. No entanto, necessita de mais pesquisas que possam relacionar os resultados obtidos neste trabalho com a produtividade da cultura em campo. O aumento da resistência à penetração não reduz a porcentagem final de emergência das plântulas de soja, mas reduz a velocidade e uniformidade de emergência. Foi observado redução da matéria seca, do comprimento total, da área superficial total e do volume total das raízes, além de maior espessamento radicular com o aumento da resistência à penetração. As respostas ao aumento da impedância mecânica na emergência e crescimento das plântulas, para a cultivar 1 e cultivar 2, foram independentes do vigor das sementes. A biossíntese do etileno não foi afetada pela impedância mecânica e vigor de sementes nas plântulas de soja aos sete dias após a emergência. O aumento da resistência à penetração não altera o conteúdo de proteína do sistema radicular e da parte aérea das plântulas da cultivar 1 e cultivar 2, mas proporcionou aumento da atividade da catalase (CAT) na raiz, aumento e redução da peroxidase do ascorbato (APX) e superóxido dismutase (SOD), respectivamente, na parte área da cultivar 1. Já a cultivar 2 apresentou maior atividade da SOD na raiz, sem alterações na atividade das demais enzimas. O lote de alto vigor da cultivar 1 tem maior expressão da enzima CAT na raiz e SOD na parte aérea das plântulas sob elevada impedância mecânica.

Mechanical impedance is considered an important concern of modern agriculture. The establishment of plants in the field depends on good emergence and seedling growth, as well as vigorous seeds. However, further research is needed to elucidate the effects of soil physical impedance on soybean seedlings growth, as well as the interference of seed vigour in the tolerance to soil compaction. In addition, it is necessary to develop more reliable methodologies for the study of soil mechanical impedance and the changes on plants caused by this stress. Thus, the objectives of this work were: i) to develop a methodology for the study of the mechanical impedance in the emergence and initial growth of soybean seedlings; ii) to evaluate the physiological and biochemical changes in soybean seedlings due to the increase in the mechanical impedance of the substrate; iii) to evaluate the effect of seed vigour on seedling tolerance to the increase in the mechanical impedance of the substrate. A system has been developed that can be used to evaluate the responses of seeds and seedlings to the physical impedance during seed germination, emergence and seedling growth. In order to validate the system, seeds of four soybean cultivars were used. The seeds were tested as for seedling emergence under different levels of mechanical impedance of the substrate (0.15, 0.60 and 0, 87 MPa), and the seedlings were evaluated for morphological characteristics. In order to evaluate the effects of the increase in the mechanical impedance of the substrate in the emergence and in the initial growth of soybean seedlings, as well as the biochemical characteristics (ethylene production, protein concentration and enzyme activities of the antioxidant system), two cultivars were used, being two lots of each cultivar, with two levels of seed vigour (high and low). Seeds were submitted to different levels of resistance to penetration of the substrate (0.15, 0.60 and 0.87 MPa) during the germination and emergence of the seedlings using the developed system. The number of emerged seedlings was evaluated daily. After seven days, the seedlings were collected and submitted to the evaluation of growth parameters (length, root diameter and dry mass), besides biochemical evaluations. The data were used to characterize the effects of the mechanical impedance on the morphological, physiological and biochemical characteristics of the seedlings. The methodology is promising for research on plant breeding aiming to verify the genotypic variability and to select genotypes more tolerant to soil compaction. However, it is necessary further research that could relate the results obtained with this work to the plant productivity on field conditions. Increasing penetration resistance of the substrate does not reduce the final emergence percentage of soybean seedlings but reduces the speed and uniformity of emergence. It was observed reduction of root dry matter, total root length, total root surface area and total root volume, as well as greater root thickening with increased resistance to penetration. The responses to mechanical impedance increasing in seedling emergence and growth, for the cultivar 1 and cultivar 2, were independent of seed vigour. Ethylene biosynthesis was not affected by substrate mechanical impedance and seed vigour in soybean seedlings seven days after emergence. The increase in resistance to penetration did not alter the protein content of the seedling root system and of the seedling shoots of the cultivar 1 and cultivar 2, but it was observed increasing in the activity of catalase (CAT) in the root, as well as increasing and reduction of ascorbate peroxidase (APX) and superoxide dismutase (SOD), respectively, in the seedling shoots of the cultivar 1. However, the cultivar 2 presented higher SOD activity in the root, without altering the activity of the other enzymes due the increasing in the mechanical impedance of the substrate. The high vigour lot of the cultivar 1 had higher expression of the CAT enzyme in the seedling root, and SOD in the seedling shoot under high mechanical impedance.
