On the track of virus yellows resistance in Beta vulgaris - molecular tools and resistance mechanisms for disease management

Der Krankheitskomplex der virösen Vergilbung [engl. Virus yellows disease (VY)] in Zuckerrüben (Beta vulgaris subsp. vulgaris L.) stellt seit dem Verbot der neonicotinoiden Saatgutbeizung ein zunehmendes Problem für den europäischen Rübenanbau dar, da sie durch verschiedene blattlausübertragbare Viren verursacht wird. Namentlich zählen zu der VY, das beet yellows virus (BYV, Gattung Closterovirus), das beet mild yellowing virus (BMYV) und das beet chlorosis virus (BChV) (beide Gattung Polerovirus), ebenso wird häufig zusätzlich das beet mosaic virus (BtMV, Gattung Potyvirus) übertragen, welches aus diesem Grund als assoziiertes Virus gilt. Ziel dieser Arbeit war es, eine rezessive Resistenz gegenüber den in Europa auftretenden Vertretern der Gattung Polerovirus als auch BtMV zu identifizieren. Als Ausgangspunkt wurde hierfür die Eigenschaft genutzt, dass sowohl Poleroviren als auch Potyviren an ihrem 5'-genomischen Ende ein kovalent gebundenes virales Protein [engl. viral protein genome linked (VPg)] tragen, das als mRNA-Cap-Analogon für die virale Translationsinitiation fungiert. Frühere Arbeiten konnten bereits zeigen, dass potyvirale VPgs mit verschiedenen eukaryotischen Translationsinitiationsfaktoren (eIFs) ihrer jeweiligen Wirtspflanzen interagierten und die Translation des viralen Genoms initiieren. Wenn die VPg-eIF-Interaktion unterbunden wird, beispielsweise durch einen homozygoten Knockout oder spezifische Mutationen, spricht man von einer rezessiven Resistenz. Die Pflanze ist infektionsresistent, da keine virale Translation erfolgen kann. Im Rahmen dieser Arbeit wurden mit Hilfe eines Hefe-Zwei-Hybrid-Systems (YTH) und der bimolekularen Fluoreszenzkomplementierung (BiFC) verschiedene Zuckerrüben-eIF-Isoformen auf ihre Interaktion mit BtMV-, BMYV- und BChV-VPg untersucht. Alle untersuchten VPgs zeigten eine Interaktion mit dem Zuckerrüben Bv-eIF(iso)4E in BiFC und YTH. Interessanterweise zeigte das BMYV-VPg in BiFC und YTH eine zusätzliche Interaktion mit der funktionell redundanten Isoform Bv-eIF4E. Nachdem die potenziellen Interaktionspartner identifiziert wurden, wurden verschiedene T0-Knockout-Zuckerrüben Linien der verschiedenen eIFs erzeugt und Resistenztests gegen die verschiedenen Virusspezies unterzogen. Durch den Knockout des Bv-eIF(iso)4E konnte eine Resistenz gegenüber BChV erzielt werden, die den Virustiter im Vergleich zur nicht editierten Zuckerrübe signifikant verringert. Sowohl für BtMV als auch BMYV konnte jedoch keine vergleichbare Resistenz gefunden werden. Die multiplen Interaktionen vom BMYV-VPg mit Bv-eIF(iso)4E und Bv-eIF4E sowohl in planta als auch in vitro, geben erste Hinweise darauf, dass möglicherweise ein simultaner Knockout der jeweiligen eIF4Es erforderlich ist, um eine Resistenz gegenüber BMYV zu erreichen. Allerdings ist davon auszugehen, dass der gleichzeitige Knockout beider eIFs für die Zuckerrübe letal ist. Daher ist die Identifizierung von eIF-Mutanten, die die mRNA-Translation ermöglichen, aber gleichzeitig nicht an das virale VPg binden, für die künftige Resistenzzüchtung erforderlich. Nichtsdestotrotz ist eine der Haupterkenntnisse dieser Arbeit, dass eine rezessive Resistenz gegenüber Poleroviren in Zuckerrüben grundlegend implementierbar ist. Obwohl die eIF vermittelte rezessive Resistenz hauptsächlich für Potyviren beschrieben ist, konnte trotz der gefundenen Interaktion zwischen BtMV-VPg und Bv-eIF(iso)4E keine Resistenz beobachtet werden. Aus diesem Grund wurden im zweiten Teil der Arbeit weitere Untersuchungen zur züchterischen Verbesserung der BtMV-Resistenz durchgeführt. Hierfür wurde der erste cDNA-Volllängenklon von BtMV hergestellt, welcher über Agrobacterium tumefaciens vermittelte Expression in Zuckerrübe infektiös ist und mit Fluoreszenz Reportergenen markiert wurde. Zusätzlich wurde ein neues Reportergen-System unter Verwendung des BvMYB1 Transkriptionsfaktors entwickelt. Durch die Verwendung von BvMYB1 wird die Betalain Biosynthese in Rüben angeregt und es entsteht eine für das bloße Auge sichtbare rote Pigmentierung im Blatt überall dort, wo eine aktive Virusreplikation stattfindet. Da Virusgehalte und Pigmentierung stark miteinander korrelieren, ermöglicht dies für die Zukunft eine schnelle und objektive Evaluierung von Symptomausprägungen in Rüben, zur Prüfung auf potentielle Resistenz. Im dritten Teil dieser Arbeit wurde unter Verwendung der neu entwickelten cDNA-Volllängenklone das bereits vor 50 Jahren beschriebene BtMV Resistenzgen Bm untersucht, da es bisher noch keine Verwendung in aktuellen Zuckerrüben Hybriden findet. Erstmalig konnte gezeigt werden, dass die Virusgehalte in Pflanzen mit dem Bm-Gen im Vergleich zur anfälligen Kontrolle signifikant verringert sind. Ebenso wird die Symptomausprägung in Bm-Pflanzen beeinflusst und mikroskopische Untersuchungen deuten auf eine verlangsamte Virusausbreitung hin. Somit konnte festgestellt werden, dass das Bm-Gen weiterhin eine effektive Möglichkeit zur Kontrolle von BtMV darstellt. Zusammenfassend lässt sich sagen, dass im Rahmen dieser Arbeit für drei von vier VY-Mitgliedern (bzw. assoziierten Viren), potentielle Kontrollmöglichkeiten für die Zukunft aufgezeigt werden konnten. Ebenso wird ein molekulares Werkzeug zur Unterstützung der Resistenzzüchtung in Zuckerrüben durch das neu entwickelte BvMYB1-Reportersystem geliefert.

The virus yellows (VY) diseases complex in sugar beet (Beta vulgaris subsp. vulgaris L.) has been an increasing problem for European beet cultivation since the ban on neonicotinoid seed treatment as it is caused by various aphid-transmissible viruses. These are namely, beet yellows virus (BYV, genus Closterovirus), beet mild yellowing virus (BMYV) and beet chlorosis virus (BChV) (both genus Polerovirus), additionally the non-yellowing beet mosaic virus (BtMV, genus Potyvirus) is also transmitted and therefore considered to be a VY-associated virus. The aim of this work was to identify a recessive resistance to the representatives of the genus Polerovirus and BtMV occurring in Europe. The starting point for this was the fact that both poleroviruses and potyviruses carry a covalently linked viral protein [viral protein genome linked (VPg)] at their 5'-genomic end, which functions as a mRNA cap analogue for viral translation initiation. Previous works have already shown that potyviral VPgs interact with various eukaryotic translation initiation factors (eIFs) of their respective host plants and initiate the translation of the viral genome. If the VPg-eIF interaction is perturbed, for example by a homozygous knockout or specific mutations, this is referred to as recessive resistance. The plant is resistant to infection because no viral translation can occur. In this work, different sugar beet eIF isoforms were investigated for interaction with BtMV-, BMYV- and BChV-VPgs using a yeast two-hybrid (YTH) system and bimolecular fluorescence complementation (BiFC) assay. All VPgs examined showed interaction with sugar beet Bv-eIF(iso)4E in BiFC and YTH. Interestingly, the BMYV-VPg showed an additional interaction with the functionally redundant isoform Bv-eIF4E in BiFC and YTH. After the potential interaction partners were identified, several T0 knockout sugar beet lines of the different eIFs were generated and subjected to a resistance test against the different virus species. Knockout of Bv-eIF(iso)4E resulted in resistance to BChV, significantly reducing the virus titer in comparison to non-edited sugar beet. No comparable resistance was found for either BtMV or BMYV. The multiple interactions of BMYV-VPg with Bv-eIF(iso)4E and Bv-eIF4E, both in planta and in vitro, give first indications that a simultaneous knockout of the respective eIF4Es may be necessary to achieve resistance against BMYV. However, simultaneous knockout of both eIF4Es is expected to be lethal for sugar beet. Therefore, the identification of eIF mutants that allow mRNA translation but at the same time do not bind to the viral VPg is necessary for future resistance breeding. Nevertheless, one of the main findings of this work is that recessive resistance against poleroviruses is implementable in sugar beet. Although eIF-mediated recessive resistance is mainly described for potyviruses, no resistance could be observed, despite the previously found interaction between BtMV-VPg and Bv-eIF(iso)4E. For this reason, further investigations were carried out in the second part of this work to improve breeding progress to generate BtMV resistance. For this purpose, the first cDNA full-length clone of BtMV was produced, which was infectious in sugar beet via Agrobacterium tumefaciens-mediated expression and was labelled with fluorescent reporter genes. In addition, a new reporter gene system was developed using the BvMYB1 transcription factor. By using BvMYB1, betalain biosynthesis in beet is stimulated and a red pigmentation visible to the naked eye develops in the leaf wherever active virus replication takes place. As virus levels and pigmentation strongly correlate, this molecular tool will allow a rapid and objective evaluation of symptom expression in beets when screening for resistance in the future. In the third part of this work, the BtMV resistance gene Bm, which was described already 50 years ago, was investigated using the newly developed full-length cDNA clones, as the gene has not yet been used in current sugar beet hybrids. For the first time, it could be shown that the virus levels in plants carrying the Bm gene are significantly reduced compared to the susceptible control. Likewise, symptom expression is influenced in Bm plants and microscopic examinations indicate a slowed virus spread. Thus, the Bm gene was found to remain an effective way for BtMV control. In summary, for three out of four VY members (or associated viruses), potential control options for the future have been identified and discussed in this work. In addition, a molecular tool to support resistance breeding in sugar beet using the newly developed BvMYB1 reporter system is provided.
