Molecular analysis of genes acting in fruiting body development in basidiomycetes

Aufgrund seines kurzen Lebenszyklus, der Fähigkeit, auf Nährmedien unter Laborbedingungen zu wachsen und Fruchtkörper zu bilden, und des sequenzierten Genoms ist Coprinopsis cinerea ein exzellentes Modell für höhere Basidiomyzeten. Diese Arbeit konzentriert sich auf Gene des Pilzes in der Fruchtkörperentwicklung. Die sexuelle Entwicklung von C. cinerea wird von den A- und B-Kreuzungstyp-Genen reguliert, die für zwei Typen von Homeodomain-Transkriptionsfaktoren (HD1 und HD2), bzw. für Pheromone und Pheromon-Rezeptoren kodieren. Selbst-fertile Mutanten wie Homokaryon AmutBmut mit defekten Kreuzungstyp-Genen werden zur Produktion von Mutanten im Fruchten benutzt.Vom Homokaryon AmutBmut wurden co-isogene Stämme mit unterschiedlichen Wildtyp-Kreuzungstyp-Spezifitäten durch wiederholte Rückkreuzungen konstruiert. Die Qualität der co-isogenen Stämme wurde mittels RAPD (Random Amplified Polymorphic DNA) Markern überprüft. Ein co-isogener Stamm wurde benutzt, um Mutantengene in der Fruchtkörperentwicklung in Kreuzungen zu verfolgen. Dadurch konnten drei verschiedene Mutationen (cfs1, mat und bad) in der AmutBmut UV-Mutante 6-031 unterschieden werden. Das Gen für eine Cyclopropan-Fettsäure-Synthase (cfs1) agiert in der Initiation von Fruchten. Die beiden anderen Mutationen befinden sich in einem Gen für Fruchtkörperprimordienreifung (mat) und in einem Gen für Basidiosporenproduktion (bad). Stamm OU3-1 mit der cfs1-Mutation und den mutierten Kreuzungstypgenen wurde für die Analyse der essentiellen cfs1-Funktion in der Fruchtkörperentwicklung mittels Transformation mit dem klonierten cfs1-Wildtyp-Gen benutzt.Die Existenz eines Anull Stammes (NA2) zeigt, dass die A-Kreuzungstyp-Gene nicht essentiell für das Wachstum sind. Während des Wachstums von NA2 Transformanten im Dunklen reprimieren eingefügte kompatible A-Kreuzungstypgene unterschiedlicher Spezifitäten die asexuelle Sporenproduktion (Oidiation). Es gibt auch Hinweise, dass auch einzelne Homeodomain-Transkriptionsgene (HD1) eine reprimierende Wirkung auf die Oidiation haben könnten.Artfremde Homeodomain-Transkriptionsfaktoren der Basidiomyzeten Ustilago maydis, Schizophyllum commune und Coprinellus disseminatus können offenbar mit denen von C. cinerea interagieren, da in C. cinerea Transformanten Schnallenbildung beobachtet wurde. Durch Transformation von B-Kreuzungstyp-ähnlichen Genen von C. disseminatus in C. cinerea konnte ferner gezeigt werden, dass die artfremden Pheromon- und Pheromon-Rezeptor-Gene bei der Schnallenfusion als auch bei der Regulation der Fruchtkörperentwicklung funktionierten.Weiter wurde ein dominant konstitutiv aktiviertes ras-Allel (rasVal19) für eine kleine GTPase, die in verschiedenen Signalwegen und eventuell auch im B-Kreuzungstyp-Signalweg involviert ist, in verschiedene C. cinerea Stämme transformiert. Der Regulator RasVal19 beeinflusste Wachstum von Mono- und Dikaryen, Gewebebildung in Fruchtkörpern und Basidiosporenproduktion.Aktivitäten der Promotoren der in S. commune im Myzel, bzw. in Fruchtkörpern exprimierten Hydrophobin-Gene Sc3 und Sc4 wurden mit einem Laccase-Gen als Reporter in C. cinerea untersucht. Beide zeigten Expression in allen Entwicklungsstadien von C. cinerea, jedoch mit Präferenz des Sc3-Promotors im Myzel und des Sc4-Promotors in den Fruchtkörpern.

Coprinopsis cinerea is an excellent model for studying fruiting body development in the higher basidiomycetes because it has a short life cycle, grows and fruits on artificial media under laboratory conditions, and in addition its genome has been released. This thesis is focused on the action of genes in fruiting body development in C. cinerea. Sexual development is regulated by the A and B mating type genes that encodes two types of homeodomain transcription factors (called HD1 and HD2) and pheromones and pheromone receptors, respectively. Self-fertile strains with defects in the mating type genes such as homokaryon AmutBmut are used to create mutants in the fruiting pathway.Co-isogenic strains with different wild type mating type specificities have been constructed from homokaryon AmutBmut by repetitive backcrosses. The quality of the obtained co-isogenic strains was tested by molecular RAPD (Randomly Amplification Polymorphism DNA) markers. A co-isogenic strain has been used to follow up in a genetic cross mutant genes in fruiting body development. In this way, three different mutations (cfs1, mat and bad) in the AmutBmut UV-mutant 6-031 could be separated. Gene cfs1 for a cyclopropane fatty acid synthase acts in the initiation of fruiting body development. The other mutations are in a gene for fruiting body primordia maturation (mat) and in a gene for basidiospore production (bad). Strain OU3-1 containing the cfs1 mutation and the defective mating type genes served further in analyzing the essential cfs1 function in fruiting development by transformation with the cloned wild type gene.The existence of an Anull strain (NA2) demonstrates that the A mating type genes are not essential for growth. However, during colony growth of NA2 transformants in the dark, introduced compatible A mating type genes of different specificities repress asexual spore production (oidiation). Some evidence is given that there might be also a reaction of single homeodomain transcription genes (HD1) on repression of oidiation.Foreign homeodomain transcription factors from the basidiomycetes Ustilago maydis, Schizophyllum commune and Coprinellus disseminatus are able to interact with homeodomain transcription factors of C. cinerea, as judged from mating-type induced clamp cell production on monokaryotic transformants of C. cinerea. In this thesis, by transformation of B mating type like genes from C. disseminatus into C. cinerea it could also be shown that the foreign pheromone and pheromone receptor genes act in clamp cell fusion and in regulation of fruiting body development.As a further regulator of development, a dominant constitutively activated ras allele (rasVal19) for a small GTPase active in different signalling pathways possibly including the B mating type pathway has been transformed into different monokaryons of C. cinerea. RasVal19 affected growth rates of monokaryons and dikaryons, tissue formation during fruiting body development, and basidiospore production.The promoters of the Sc3 and Sc4 hydrophobin genes of S. commune, two developmentally regulated genes expressed in vegetative mycelium and fruiting bodies, respectively, were studied in activity in C. cinerea, making use of a laccase gene as a reporter. Expression from the two promoters were found in all development stages in C. cinerea but with a preference for the Sc3 promoter in the vegetative mycelium and the Sc4 promoter in the fruiting bodies.
