Einfluss von Enterococcus faecium als Probiotikum auf Salmonella Typhimurium DT104 am Infektionsmodell Schwein | Influence of a probiotic strain of Enterococcus faecium on Salmonella Typhimurium DT104 infection in swine as animal model

Obwohl die fördernde nutritive Wirkung der probiotischen Enterococcus spp. auf Mäuse und den Menschen bereits beschrieben wurde, sind experimentelle Studien bei Grosstieren als auch Challenge-Versuche mit pathogenen Mikroorganismen selten. In dieser Arbeit wurde der Einfluss der oralen Gabe von Enterococcus faecium NCIMB 10415 auf mit Salmonella Typhimurium DT104 infizierte Absetzverkel untersucht. Zu unterschiedlichen Zeitpunkten nach dem Challenge (3, 24, 72 Stunden und 28 Tage p.i.) wurden die klinischen Symptome, die Ausscheidung im Kot und die Verteilung der Salmonellen in ausgewählten Organen als auch die humorale Immunantwort (anti-Salmonella-IgG, -IgM und –IgA) im Serum der Ferkel analysiert. Insgesamt wurden 89 Absetzferkel zufällig in die Probiotika- (43 Tiere) und Kontrollgruppe (46 Tiere) eingeteilt. Die Tiere der Probiotikagruppe hatten Kontakt mit Enterococcus faecium sekundär über die Muttersau (Kot) und primär durch das Futter ab dem 14. Lebenstag. Nach dem Challenge mit Salmonella typhimurium DT104 zeigten die Ferkel beider Fütterungsgruppen keine schweren klinischen Symptome einer Salmonellose. Allerdings war die Ausscheidung im Kot und die Besiedlung der Organe mit Salmonellen signifikant höher bei den mit Enterococcus faecium gefütterten Ferkeln. Gleichermaßen war die humorale Immunantwort gegen Salmonellen (spezifisches Serum-IgM und -IgA) signifikant höher in der Probiotikagruppe im Vergleich zu den Kontrolltieren. Nach den Ergebnissen dieser Studie führte die Gabe von Enterococcus faecium NCIMB 10415 bei den Absetzferkeln der Probiotikagruppe zu einem geringeren Schutz nach Challenge mit Salmonella typhimurium DT104 als bei den Tieren der Kontrollgruppe, was mit der erhöhten Ausscheidung der Salmonellen im Kot und Manifestation in den Organen als auch anhand der stärkeren Produktion spezifischer Antikörper gegen Salmonella Typhimurium DT104 bei den Tieren der Probiotikagruppe gezeigt werden konnte. Ob die humoralen Mechanismen, d.h. IgA, IgM und/oder IgG Titer, eine verbesserte Immunantwort auf die Probiotikabehandlung darstellen oder eine natürliche Immunantwort auf die erhöhte Salmonellenbelastung in den Organen sind, muss durch weitere Untersuchungen geklärt werden.

Although beneficial nutritive effects of probiotic Enterococcus spp. on different hosts such as mice and humans have previously been reported, experimental studies on large domestic animals, as well as challenge studies with pathogenic microorganism are very rare. In this study, the influence of oral treatment of pigs with the probiotic bacterium Enterococcus faecium NCIMB 10415 on Salmonella Typhimurium DT104 infections in weaning piglets has been investigated. At different periods after challenge (3, 24, 48 h and 28 days p.i.), clinical symptoms, fecal excretion, the distribution of Salmonella in selected organs, as well as the humoral immune response (Anti-Salmonella-IgG, -IgM, and -IgA) in serum of piglets was analysed. A total of 89 piglets was randomly divided into the probiotic (43 animals) and control group (46 animals). The animals of probiotic group had contact to Enterococcus faecium secondarily through the mother sow (milk, faeces) and primarily through the supplemented feed, starting from day 14 post partum. After challenge with Salmonella Typhimurium DT104, piglets of both groups showed no severe clinical signs of salmonellosis. However, fecal excretion and colonization of Salmonella in organs was significantly higher in piglets fed with E. faecium. Likewise, the humoral immune response against Salmonella (specific serum IgM and IgA) was significantly higher in the probiotic group compared to control animals. Results of this study suggest that application of Enterococcus faecium NCIMB 10415 enhanced the course of infection in weaning piglets challenged with Salmonella Typhimurium DT104, which was shown by the increased excretion of Salmonella in faeces and manifestation in internal organs as well as the higher production of specific antibodies against Salmonella Typhimurium DT104 in animals of the probiotic group. Whether the humoral mechanisms, i.e. IgA, IgM and/or IgG titers, represented an enhanced immune response as a result of the probiotic treatment or the natural immune response to elevated Salmonella loads in organs will require further study.
