Anthelmintic resistance in intestinal parasites of dogs in north and center of Portugal

Os ancilostomatídeos, os ascarídeos, os tricurídeos e os parasitas da família Taeniidae são bem conhecidos como sendo os parasitas intestinais mais prevalentes em cães. A redução da contagem de ovos fecais nas fezes é um método amplamente utilizado para avaliar a eficácia de anti-helmínticos contra parasitas gastrointestinais. Os objectivos deste estudo foram: realizar um estudo epidemiológico sobre diversidade e carga parasitária de helmintes intestinais e avaliar possíveis resistências a anti-helmínticos, em cães das regiões Norte e Centro de Portugal. Para o efeito foram recolhidas amostras fecais de cães em 18 concelhos, provenientes de 14 centros de recolhas oficiais, 5 canis de associações privadas e de 11 tutores particulares. Estas amostras foram analisadas através da metodologia Mini-FLOTAC®, em dois tempos distintos, no dia ou na véspera das desparasitações e 13 dias após as mesmas. A percentagem de eficácia foi calculada usando a fórmula das diretrizes da World Association for the Advancement of Veterinary Parasitology (WAAVP), e considerou-se que o anti-helmíntico era ineficaz, de baixa eficácia e moderadamente eficaz, se a redução de carga parasitária fosse menor de 60%, entre 60-79% e entre 80-89%, respetivamente. Para a análise estatística foi usado o programa R. Usou-se o teste qui-quadrado de Pearson para dados categóricos. Os valores P < 0,05 foram considerados estatisticamente significativos. Das 361 amostras fecais de cães recolhidas, foram identificadas formas parasitárias em 22,4%, nomeadamente os nematodes Trichuris vulpis (10,5%), Toxocara canis (8,0%), Ancylostomatidae (6,4%) e Toxascaris leonina (0,8%), e os cestodes Dipylidium caninum (0,6%) e Taeniidae (0,3%). Os primeiros três parasitas nematodes tiveram uma elevada intensidade de infeção em alguns cães, com uma eliminação máxima de 2900, 1325 e 1265 ovos por grama, respetivamente. Treze dias após tratamento antihelmíntico, foi realizada uma segunda colheita e análise fecal dos cães infetados na primeira recolha (n=77). Na segunda recolha, 20,8% das amostras evidenciaram formas parasitárias. De acordo com a percentagem de redução da carga parasitária verificou-se que para Trichuris vulpis, os anti-helmínticos febendazol + embonato de pirantel + praziquantel e praziquantel + fenbendazol foram ineficazes em 13,5% das amostras (n=5), febantel + praziquantel + pirantel e febendazol + embonato de pirantel + praziquantel tiveram baixa eficácia em 5,4% (n=2) e praziquantel + fenbendazol foi moderadamente eficaz em 2,7% (n=1). Já para Ancylostomatidae febendazol + embonato de pirantel + praziquantel e praziquantel + fenbendazol foram ineficazes em 6,8% (n=2) e febendazol + embonato de pirantel + praziquantel foi também moderadamente eficaz em 3,3% (n=1). As prevalências (geral e por forma parasitária) deste estudo não foram elevadas quando comparadas com outros realizados anteriormente em Portugal, contudo as formas parasitárias identificadas são todas reconhecidamente ou potencialmente zoonóticas, além de causadoras de doença animal. Este estudo é o primeiro a investigar a resistência anti-helmíntica em parasitas intestinais de cães em Portugal e a identificar ineficácia dos anti-helmínticos para Trichuris vulpis e Ancylostomatidae. Os resultados de baixa eficácia obtidos em poucos hospedeiros não sugerem resistência da comunidade de parasitas intestinais, mas podem indicar o metabolismo deficiente dos hospedeiros, pobre viabilidade do fármaco, reinfeção, entre outros fatores. Outros estudos deverão ser realizados no futuro para entender a extensão da resistência anti-helmíntica.

Hookworms, roundworms, whipworms and parasites of the Taeniidae family are well known to be the most prevalent intestinal parasites in dogs. Reducing faecal egg counts in faeces is a widely used method to assess the effectiveness of anthelmintics against gastrointestinal parasites. The aims of this study were: to carry out an epidemiological study on intestinal helminths diversity and parasite burden and to assess possible resistance to anthelmintics in dogs from the North and Center regions of Portugal. For this purpose, faecal samples were collected from dogs in 18 municipalities, from 14 public shelter dogs, 5 private shelter dogs and 11 private tutors. These samples were analysed using the Mini-FLOTAC® methodology, at two different times, on the day or the day before the deworming and 13 days after the same. Percentage of efficacy was calculated using the World Association for the Advancement of Veterinary Parasitology (WAAVP) guidelines formula, and the anthelmintics were considered to be ineffective, of low efficacy, and moderately effective if the reduction in parasite load was less than 60%, between 60-79% and between 80-89%, respectively. For statistical analysis, the R program was used. Pearson's chi-square test was used for categorical data. The values P < 0.05 were considered statistically significant. Of the 361 faecal samples collected from dogs, parasitic forms were identified in 22.4%, such as the nematodes Trichuris vulpis (10.5%), Toxocara canis (8.0%), Ancylostomatidae (6.4%) and Toxascaris leonina (0.8%), and the cestodes Dipylidium caninum (0.6%) and Taeniidae (0.3%). The first three nematode parasites had a high infection intensity in some dogs, with a total egg shedding of 2900, 1325 and 1265 egg per gram, respectively. Thirteen days after anthelmintic treatment, a second collection and faecal analysis was performed on infected dogs in the first collection (n=77). In the second collection, 20.8% of the samples showed parasitic forms. According to the percentage of reduction in the parasite load, it was found that for Trichuris vulpis, the anthelmintics febendazole + pyrantel embonate + praziquantel and praziquantel + fenbendazole were ineffective in 13.5% of the samples (n=5), febantel + praziquantel + pyrantel and febendazole + pyrantel embonate + praziquantel had low efficacy in 5.4% (n=2) and praziquantel + fenbendazole was moderately effective in 2.7% (n=1). For Ancylostomatidae, febendazole + pyrantel embonate + praziquantel and praziquantel + fenbendazole were ineffective in 6.8% (n=2) and febendazole + pyrantel embonate + praziquantel was also moderately effective in 3.3% (n=1). The overall and by parasite prevalences of thisstudy were not high when compared to others previously carried out in Portugal, however the parasitic forms identified are all recognizably or potentially zoonotic, in addition to causing animal disease. This study is the first to investigate anthelmintic resistance in intestinal parasites of dogs in Portugal and to identify ineffectiveness of anthelmintics for Trichuris vulpis and Ancylostomatidae. The low efficacy results obtained in few hosts do not suggest resistance of the intestinal parasite community, but may indicate deficient host metabolism, poor drug viability, reinfection, among other factors. Further studies should be carried out in the future to understand the extent of anthelmintic resistance.
