Production of ochratoxin A, fumonisins and emodin by Aspergillus niger isolates from feed products | Продуцирование охратоксина А, фумонизинов и эмодина у вида Aspergillus niger из кормовой продукции

Micromycetes of one of the most complex taxonomically Aspergillus niger complex (the A. niger "aggregate") have invariably remained the object of close attention of researchers due to the risks of negative effects on humans and animals. The study purpose was to assess the ability of A. niger cultures isolated from compound feed, five types of feed raw materials and hay of different botanical composition and territorial origin to produce B group fumonisins (FUM), ochratoxin A (OA), emodin (EMO), as well as to clarify their species according to the totality of morphological, physiological characteristics and composition of metabolites. For 12 monoconidial strains, toxin production was assessed on sucrose agar with yeast extract (YES), Czapek agar with yeast autolysate extract and 20% sucrose (CYA20S) and on rice grain. After incubation (7 days, 25 deg. C), the extracts were analyzed using commercial domestic test systems for mycotoxin determination by ELISA developed at the All-Russian Research Institute of Veterinary Sanitation, Hygiene and Ecology. According to morphological criteria, according to the absence of sclerotia, results of Ehrlich test and the ability to produce OA and FUM, their belonging to the species A. niger in the clade of biseriate species was confirmed. The accumulation of FUM in 7 strains was 0.2 to 630 mkg/g and absent in 5; all strains produced OA (from 0.005 to 0.064 mkg/g) and EMO (from 0.004 to 0.9 mkg/g). The difference in the accumulation of mycotoxins on agar media and grain substrate confirms the influence of substrate components on the activity of determinant gene clusters. Due to the weak potential for producing OA, EMO and the small proportion of active FUM producers, the contribution of A. niger to the contamination of grain feed and hay, even with intensive infestation, is unlikely to be significant.

Микромицеты одного из самых сложных в таксономическом отношении комплекса Aspergillus niger (the A. niger "aggregate") неизменно остаются объектом пристального внимания исследователей в связи с рисками негативных воздействий на человека и животных. Цель работы — оценка способности культур Aspergillus niger, выделенных из комбикорма, пяти видов комбикормового сырья и сена разного ботанического состава и территориального происхождения, продуцировать фумонизины группы В, охратоксин А и эмодин, а также уточнение их видовой принадлежности по совокупности морфологических, физиологических характеристик и составу метаболитов. Оценивали токсинообразование 12 моноконидиальных культур, растущих на сахарозном агаре с дрожжевым экстрактом (YES), агаре Чапека с экстрактом автолизата дрожжей и 20 % сахарозы (CYA20S) и на зерне риса. После инкубирования (7 сут, 25 град. С) анализ экстрактов выполняли с помощью тест-систем для иммуноферментного определения микотоксинов (СТО ВНИИВСГЭ, Россия). По морфологическим критериям, по отсутствию склероциев, результату теста Эрлиха и способности продуцировать ОА и ФУМ подтверждена принадлежность культур виду A. niger в кладе двухъярусных видов. Накопление ФУМ у 7 штаммов составляло 0,2 до 630 мкг/г и у 5 отсутствовало, все штаммы продуцировали ОА (от 0,005 до 0,064 мкг/г) и ЭМО (от 0,004 до 0,9 мкг/г). Различие в накоплении микотоксинов на агаровых средах и зерновом субстрате подтвердило влияние компонентов субстратов на активность детерминирующих кластеров генов. Из-за слабого потенциала продуцирования ОА, ЭМО и небольшой доли активных продуцентов ФУМ вклад A. niger в контаминацию зерновых кормов и сена (даже при интенсивной пораженности) вряд ли может быть существенным.
