Thermal inactivation mode of myofibrillar Ca-ATPase of kuruma prawn Marusupenaeus japonicus

The thermal stability of kuruma prawn myofibrils (Mf) was investigated using myofibrillar Ca-ATPase as an index. Heat treatment of Mf caused inactivation of myofibrillar Ca-ATPase with a biphasic first order process. The inactivation rate constants in the earlier and the later phases were similar to those of walleye pollock and rainbow trout Mf, respectively. Moreover, the biphasic first order process of the thermal inactivation did not change with the addition of F-actin to Mf prior to heat treatment. The optimum temperature of myofibrillar Ca-ATPase activity was raised by intentionally removing the earlier phase in the thermal inactivation mode with heat treatment. These results suggest that the thermal stability of myofibrillar Ca-ATPase prepared from kuruma prawn muscle was not uniform. One portion exhibited a stable state and another exhibited a more unstable state.
