On the metabolism and toxicokinetics of deoxynivalenol and zearalenone in endotoxaemic pigs

德语. Ziel dieser Arbeit war die Erforschung der Wechselwirkung zwischen einer Mykotoxinexposition und einer Immunantwort. Es wurden zwei Aspekte untersucht: 1. Die systemische Antwort auf eine pre- oder post-hepatische Endotoxin-Challenge, unter dem Einfluss einer chronischen Deoxynivalenol (DON)- und Zearalenon (ZEN)-Fütterung; und: 2. Der Einfluss einer Endotoxin-Challenge auf die Metabolisierung von DON und ZEN. Hierfür wurde ein in vivo Versuch mit 44 männlichen Schweinen der deutschen Landrasse durchgeführt. Die Ergebnisse zeigen, dass die systemische Antwort auf eine Endotoxin-Challenge sich anhand der Expositionsroute, aufgrund des „first-pass“ Effektes der Leber, unterscheidet und dass eine chronische Mykotoxin Fütterung einen Einfluss auf diese Pathogenese hat. Des Weiteren bestätigen die Ergebnisse, dass sich unter einer akuten Phasen Reaktion die Toxikokinetik von Mykotoxinen verändert. Dies konnte vor allem für ZEN und seine Metaboliten gezeigt werden.

英语. The aim of this work was to investigate the interaction between mycotoxin exposure and an immune response. Two aspects were investigated: 1. the systemic response to a pre- or post-hepatic endotoxin challenge, under the influence of a chronic deoxynivalenol (DON) and zearalenone (ZEN) feeding; and: 2. the influence of an endotoxin challenge on the metabolization of DON and ZEN. For this, an in vivo experiment was performed including 44 male pigs of the german land race. The results show that the systemic response to an endotoxin challenge differs based on the route of exposure, due to the "first-pass" effect of the liver, and that chronic mycotoxin feeding has an impact on this pathogenesis. Furthermore, the results confirm that under an acute phase reaction the toxicokinetics of mycotoxins changes. This was especially demonstrated for ZEN and its metabolites.
