Endophytic bacteria of the genus Bacillus for protecting potato and tomato plants from early blight pathogen | Эндофитные бактерии рода Bacillus для защиты растений картофеля и томатов от альтернариоза

英语. Early blight, caused by the fungus Alternaria solani, is common in all major Solanaceae plants growing regions. The most well-known biological control agents are strains of Bacillus species (B. thuringiensis, B. subtilis and B. velezensis) found in the phyllosphere and rhizosphere of numerous crops. The work aim was to determine the ability of proteolytic enzyme-producing strains of Bacillus bacteria, isolated in the South Urals, to colonise the internal tissues of tomato and potato plants, as well as to increase the resistance of these crops to the early blight pathogen in model experiments. Two types of plants were inoculated with tested strains: sterile in vitro to count endophytic cells and grown in cups filled with soil to investigate Alternaria symptoms on leaves after artificial infection. For the first time, the accumulation of viable bacterial cells of these strains in the internal tissues of the stem and the ability of endophytic bacteria to use pectin and starch as a source of carbohydrates was demonstrated. A significant reduction in the area affected by Alternaria on the leaves of plants inoculated with endophytes was revealed, as well as inhibition of pathogen growth under the influence of bacterial strains in vitro, which can be explained by the synthesis of proteolytic enzymes. А decrease in the manifestations of Alternaria blight on potato leaves was observed under the influence of B. subtilis 26D, B. subtilis Ttl2 and B. sp Ttl1; on tomato leaves – following the application of B. subtilis 26D and B. sp Ttl1. The lowest score of early blight development was observed on leaves treated with B. velezensis M66, which was also characterized by the highest colonization activity in relation to both crops. At the same time, treatment of plants with B. sp. TV2, the cell content of which was much lower, did not lead to a significant reduction in the manifestation of disease symptoms on the leaves of both crops. Furthermore, B. subtilis Ttl2, which colonizes tomato tissue to a small extent, did not show a protective effect on tomato plants. Thus, these endophytic strains can be considered as the most promising biocontrol agents.

俄语. Цель работы - определить способность продуцирующих протеолитические ферменты штаммов бактерий р. Bacillus (B. thuringiensis, B. subtilis и B. velezensis), изолированных на территории Южного Урала, колонизировать внутренние ткани растений томатов и картофеля и их возможность увеличивать устойчивость этих культур к возбудителю альтернариоза в условиях модельных опытов. Использовали инокулированные исследуемыми штаммами стерильные пробирочные (учет эндофитных клеток) и выращенные в стаканчиках с почвой растения (исследование симптомов альтернариоза на листьях после искусственного инфицирования). Показано накопление жизнеспособных клеток бактерий этих штаммов во внутренних тканях стебля и способность эндофитных бактерий использовать пектин и крахмал в качестве источника углеводов. Выявлено значительное сокращение площади поражения альтернариозом на листьях инокулированных эндофитами растений, а также ингибирование роста патогена под действием бактериальных штаммов in vitro, что может объясняться синтезом протеолитических ферментов. Снижение проявлений альтернариоза на листьях картофеля наблюдалось под действием B. subtilis 26Д, B. subtilis Ttl2 и B. sp Ttl1, а на листьях томатов – B. subtilis 26Д и B. sp Ttl1. Самый низкий балл развития альтернариоза наблюдался на листьях, обработанных B. velezensis M66, который также характеризовался наибольшей колонизационной активностью по отношению к обеим культурам. Обработка растений B. sp. TV2, содержание клеток которого было гораздо меньшим, не привела к существенному снижению проявления симптомов заболевания на листьях обеих культур, а B. subtilis Ttl2, в малой степени заселяющий ткани томатов, не проявил защитного эффекта по отношению к растениям томата. Данные эндофитные штаммы можно рассматривать как наиболее перспективные агенты биоконтроля, что, вероятно, объясняется длительным присутствием в надземной части растения и защищенностью от воздействия окружающей среды.
